Deficiency Of Kidney Essence Of Male Mouse Liver Main Function Of Research

Objective The liver and kidney promote and reinforce each other,deficiency of the one will lead to deficiency of the other,and the physiologic and pathologic condition of the liver may be reflected in the changes of the eyes. After the hypothesis, we researched the affect of mice which has insufficient ShenJing that will lead to deficiency of the liver and the mechanism of these affects, the models were made by the "affair combined with excessive sexual behavior".Method The mice were allocated to 3 groups(blank control group, model group,Bugan group) by random number. The mice of the model group and the Bugan group were processed by the "affair combined with excessive sexual behavior" to made the model of .insufficient ShenJing. The mice in Bugan group were lavaged of concentrated solution of Yi Guan prescription, the dosage is 0.3ml/10g based on their weigh.Other groups treat with physiological saline at the same dosage.This phase would continued 21 days. And then we detect items below: i The difference of histopathology and ultramicrostructure of retinal; ii Apoptosis of the retina was assayed by both DNA fragmentation gel-electrophoresis and term inal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate-biotin nick end labelling(TUNEL); iii To reseaech the expression of aopotosis-related genes(Bcl-2, Bax) in retinas; iv The level of BDNF mRNA was determined by reverse transcription-polymerase chain reaction(RT-PCR).Result For model mice, the retinal's cells was denatured,edema,disordered,thickner.It can be found through electron microscope that the external limiting mrmbrane is edematous,unclear. All of these damage were much so lighter in Bugan groups, and some of item were normal.DNA ladder pattern was detected in model group.Further,immunochemical stain of the eye demostrated that TUNEL positive cells were localized in GCL and INL of retina,the positive reaction of model mice increased significant compared with cotrol and Bugan group(p<0.01).The express of Be 1-2 and Bax in the GCL and INL of retina is geting increased, the positive reaction of model mice increased significant compared with cotrol and Bugan group(p<0.01). Cotrol and Bugan group retinal cells expressed higer levels of BDNF mRNA than the model mice.Conclusion The deficiency of the kidney will lead to deficiency of the liver.The mechanisim of the insufficient of ShenJing lead the damage of the liver, maybe it influence the ratio of aopotosis-related genes Be 1-2 and Bax,at the same time inhibit the expresstion of the protection factor BDNF,and the apoptosis of retina is rised.These adverse effect could be prevented by Yi Guan prescription.
