Objective The males ShenJing are very important for the health of their offspring , based on the "pregnant theory" of Traditional Chinese Medicine. After this hypothesis, we researched the affect of male mice which has insufficient ShenJing on the reproductive system of their offspring and the mechanism of these affects, the models were made by the "affair combined with excessive sexual behavior".Materials and methods Materials: KunMing mice, 6 weeks old, 30 males and 330 females. Average weight is 32(s=2.5)g. The mice come from The Medicine Science academic of HuBei Province. The mice were allocated to 3 groups(blank control group, model group, BuShen group) by random number. Method: The mice of the model group and the BuShen group were processed by the "affair combined with excessive sexual behavior" to made the model of .insufficient ShenJing. The mice in BuShen group were lavaged of concentrated solution of BuShenFang, the dosage is 0.16ml/10g based on their weigh.Other groups treat with physiological saline at the same dosage. This phase would continued 21 days. All of 3 groups were matched-pairs with health female mice for 5 days after model making. And then we detect items below: i, sperm count and motility rate; ii pregnancy rate and average baby counts of every female mouse(sum of babies/number of mother mice); iii Collect sperms from epididymis, and detect sperm DNA damage by SCGE and SCSA; iv The difference of histopathology and ultramicrostructure of testis, and evaluate these difference by Johnsen Score, the diameter of seminiferous tubules and the thickenss of seminiferous epithetlium. Detect items below when the F1 mice were 6 weeks old: i sperm count and motility rate; ii The T and FSH level in serum; iii The concentration of MDA, SOD, T-AOC in testicle tissue; iv Detect the apoptosis rate of spermatogenic cell by flow cytometry; v Collect sperms from epididymis, and detect sperm DNA damage by SCGE and SCSA; vi Detect the difference of the expressive of mRNA in testis by difference display PCR; vii The difference of histopathology and ultramicrostructure of testis, and evaluate these difference by Johnsen Score, the diameter of seminiferous tubules and the thickenss of seminiferous epithetlium.Result For model mice, the Sertoli's cells wasdenatured,seminiferous tubules were atrophy in different degree, seminiferous epithetlium were thinningzed and the arrangement of cells was disordered.
