Objective: To study the mechnism of dopamine receptor (DAr) on heroin euphoria memory trace by electroacupuncture therapy.Methods: 90 male mice were be randomly divided into 6 groups after filtration by CPP, the groups werexontrary group (A), contrary with acupture group (B), model group (C), model with acupoint group (D), model with no-acupoint group (E), western medicine group (F). Heroin psychological dependence mice was formed by giving heroin with increasing dosage day by day. Mice were treated by electroacupuncture on acupoints as"Neiguan", "Sanyinjiao", "Sishengchong" once a day and lasted for 10 days. The behaviour index was measured by CPP (conditioned place preference), and dopamine receptor (D1r, D2r, D3r, D4r) and expression of DAr was be measured by immunohistochemistry (IH) and in situ hybridization histochemistry (ISH).Results: 1. mice of C group (12.97±3.11) stayed longer in CPP than which of A group (2.04±0.05) and B group (2.05±0.04), P<0.01; there has significant difference among D group, F group and C group, P<0.01; there has no difference between E group and C group, P>0.05. 2. Result of D1: result of IH: D1 expression of C group is increased, its positive cell count (88.15±3.73) and OD (97584.08±5231.47) is significant different to of A group (48.23±0.64, 31750.68±4325.7) and of B group (49.01±0.58, 32053.31±4100.25), P<0.01; D1 expression of D and F group are decreased, IH and OD of D group (60.11±4.55, 50217.54±3897.44),of F group (62.45±8.73, 55854.76±4125.56) are significant different to C group, P<0.01; there has no difference between Egroup (82.85±10.17,92397.45±4254.17) and C group, P>0.05. Result of ISH: Dl mRNA expression of C group is increased, its positive cell count (92.18±3.47) and OD (10712.14±4379.28)are significant different to which of A group (40.05±2.16, 28979.14±3012.48), of B group (41.11±2.57, 29151.86±3155.73), P<0.01; Dl mRNA expression is decreased, which is significant different to C group, PO.Oljthere has no difference between E group and C group, P<0.05. 3. Result of D2: result of IH: D2 expression of C group is increased, its positive cell count (86.24±6.23) and OD (99125.46±3478.26) are significant different to of A group (55.11±4.18, 30274.16±2988.10), of B group (56.42±5.49, 31572.39±3007.23), P<0.01; D2 exprssion of D group (66.94±6.01, 43174.14±3011.24) and of F group (66.10±8.28, 44152.08±3157.40) are decreasd , they are significant different to of C group, P<0.01; there has no different between E group (82.15±7.11, 90023.17±4012.09) and C group, P>0.05. Result of ISH: D2 mRNA expression is increased, its positive cell count (108.12±8.13) and OD (117856.27±4672.18) are significant different to of A group (52.43±6.99, 37250.84±2375.29) and of B group (56.14±5.45, 39954.25±2041.25), PO.01; D2mRNA expression of D group (60.16±7.17, 62019.18±4071.25), and of F group (62.17±8.84, 63879.18±4382.18) are decreased, they are significant different to of C group, PO.01; there-has no different between E group (104.46±9.24, 100245.44±4495.39) and C group, P>0.05. 4. Result of D3: result of IH: D3expression of C group is increased, its positive cell count (96.77±10.19) and OD (100239.17±6724.29) are significant different to of A group (52.15±3.88 , 42839.16±4973.12), of B group (54.11±3.02 , 44151.03±4425.82), P<0.01; D3 expression of D group (62.73±12.18 ,62185.29±6614.28) is decreased, which is significant different to of C group, P<0.01; there has no difference between F group (92.11±14.27 , 100120.14±5378.90), E group (95.89±16.87, 100940.28±5354.29) and C group, P>0.05. Result of ISH: D3 mRNA expression of C group is increased, its positive cell count (99.17±13.27), OD (99469.22±7821.08) are significant different to of A group (43.78±10.11 , 31459.28±4197.45), of B group (45.62±9.18, 32461.46±4218.33), P<0.01; D3 mRNA expression is decreased (66.71=fcl0.25, 56208.28±4501.25), which is significant different to of C group, PO.01; there has no difference between E group (97.12±11.29 , 94320.11±6018.24), F group (93.44±21.46, 95012.47±6110.25) and C group, P>0.05. 5. Result of D4: result of IH: D4 expression of C group is increased, its positive cell count (93.26± 15.38), OD (98124.35±3582.19) are significant different to of A group (60.15±8.73, 39756.12±3142.18) and of B group' (62.19±13.26, 40012.25±3324.15), PO.01; there has no difference between D group (91.48±16.35 , 96358.17±3818.28), E group (92.01dbl5.48 , 95290.28db44U.06), F group (90.12±24.15, 95438.10±4715.24) and C group, P>0.05. Result of ISH: D4 mRNA expression is increased , its positive cell count (112.58±14.77), OD (125024.28±10358.27) are significant different to of A group (66.01±10.12, 47320.11±4724.29), of B group (62.15±13.49, 50121.36±4971.14), P<0.01; there has no difference between D group (108.24±20.73 , 12018O.O9±l 1040.19), E group (102.15±20.19 , 123105.42±10571.27), F group (105.27±19.45, 119798.25±10298.37) and Cgroup, P>0.05.Conclusions: 1.
