| The study of "Four seasons,five zang-organs and yinyang" has gained distinct achievements in many aspects of experiments and literatures after several years directed by professor CHENG Shi-de and GUO Xia-zhen.This theory figures that human body is an organic integer,in which five zang-organs is the center and harmonizes the function or process of apparatuses with diversification of nature. Thinking of limitations of animal experiments can't compeletely open out the mechanism how different seasons and climate affect human beings'circadians and pathological changing,so we put up the research in this field.We hope that we can find out new ways in integration of clinic and the achievements in "Four seasons,five zang-organs and yinyang". 1 Objective and Methods In literature research,this study tends to probe into the "doctrine of the five circuit phases and six atmospheric influences",which is the optimum reflection on how different seasons and climate affect human beings'circadians and pathological changing in Traditional Chinese Medicine(TCM) theory. We summarize and evaluate the literature of this doctrine in past dynasties,so we can discuss its scientific value and the way to study the mechanism how different seasons and climate affect human beings'circadians and pathological changing.We also discuss relativity between weather in winter and occurrence of diseases by analyzing doctrine of the five circuit phases and six atmospheric influences. In experiments study,we find that few studies chose human being as testees instead of animals(rats),which has different behaviour compared with human beings. We also find that there is no study involved in changing rule of melatonin in different solar terms in melatonin study,so we observe the changing rule in serum or saliva samples of human being in different solar terms (spring begins,vernal equinox, summer begins, the Summer Solstice, autumn begins, autumnal equinox, winter begins,midwinter),so that we can discuss the scientific principles on how different seasons and climate affect human beings'circadians and pathological changing. There are three main methods in determining melatonin,which are Gas chromatography-Mass Spectrometry ( GCMS ), High-Performance Liquid Chromatography(HPLC)and Radioimmunoassay(RIA). GC-MS is used to determine the trustiness of other methods and HPLC has merits in mensurating similar indole compounds in one sample.But the price and service of instruments are expensive in GC-MS and HPLC which are also not suitable to mensurate large quantities of samples,we have to choose RIA as the method.As there is no melatonin RIA Kit in our country and the overseas Kits which are mostly made in America and Germany are very expensive,we develop Melatonin RIA Kit combining with higrade reseach center.Melatonin antiserum is the vital ingredient in the Kit,so we preparate melatonin antiserum.As there is some difficulties to gain serum samples from human body,we feel out the method to determine the content of melatonin in saliva samples,so that we can use saliva as samples instead of serum. Using statistical software SPSS10.0, One-way ANONA was engaged for data analysis. 2 Results 2.1 Results of literature research 2.1.1 Most scholars affirmed the scientific value of "doctrine of the five circuit phases and six atmospheric influences",and we analyze the arguments of opponents and clarify their viewpoints so that the scientific value is ensured. 2.1.2 The doctrine of the five circuit phases and six atmospheric influences is a kind of scientific deduction.Although it is based on clinics,it has not been validated adequately.In clinics,it is used to forecast climate changing and diseases prevailing approximately,so that it can help to improve the level diagnoses and therapies. 2.1.3 Forecasting may be the way to study the scientific principles on how different seasons and climate affect human beings'circadians and pathological changing by using this theory.It will be a good direction to forecast and cure infectious disease by using this...
