Osteoarthritis(OA)caused by the every kind of and different reasons is the prevalent articular disease that is characteristic of joint cartilage degeneration. It is common diseasee. It is well know that OA is due to turbulence of burthen, the low activity of the superoxide dismutase, cell factor, high intraosseous pressure and so on. Taking AA male chicken as study object because they walk on two legs like human being, we study the effect of treatment OA with external fixator which was used for reducing burthen on articular cartilage. The "GU YAN DING, GYD" which Professor Chen Ji-Chong invented can alleviate suffering of OA, promote the restoration of joint function. The ingredients of decoction of GU YAN DING are Carthamus tinctorius, Achyranthes bidentata, Psoralea corylifolia, Drynaria fortunei, Astrahalus membranaceus. The decoction have the function of nourishing the kidney and liver, replenishing qi and promoting blood circulation. In order to expound the pharmacological mechanism of GYD , author study how the decoction of GYD influence the chicken ankle articular cartilage histomorphology, and proliferation of human umbilicalvein endothelial cell.Test 1 An Observation of the Pathology of Chicken Knee OA Treated with External Fixator and Decoction of GYDObjective: The chicken knee joint was taken as our study objective, we study how the external fixator and decoction of GYD influence the pathology of chicken knee OA. Method: Adopted 25 male chickens of 3 months old, these chickens were randomly divide into 5 groups. There were 5 chickens in control group . The knee OA were established by cutting off posterior cruciate ligament and anterior cruciate ligament and menisci in OA model group. The operation was successful if the anterior drawer test was positive . In GYD group , the chickens after operation of OA model were fed decoction of GYD. In external fixator group , the chickens knee afer operation of OA model were treated with external fixator for reducing burden of articular surface .In external fixator and GYD group , the chickens operated were treated with external fixator and fed decoction . Excised the cartilage of femur after twelve weeks for the observation by photo-microscope. Result: Obviously degenerative changes were found in the OA model group. The knee joint's soft tissue of OA model group were found adhesion, overgrowth, especially at distal joint cartilage surface, which can protect distal cartilage. The articular cartilage surface were found rough, soft, ulcer, osteophytes. The cartilage cell were found disappeared and shrinked by microscopy. It is successful that chicken was used for knee OA model as experimental animal. The degree of degenerative changes in OA model group was higher than in decoction of GYD group. The degree of degenerative changes in both external fixator groups and in OA model group was not differentobviously. Conclusion: This kind of external fixator can not protect articular cartilage, the decoction of GYD can protect articular cartilage.Test! The effect of GYD on SOD and MDA of chicken knee joint OA Objective: Study how the decoction of GYD influence the activity of the superoxide dismutase of chickens OA model for pharmacological mechanisms of decoction of GYD. Method: 40 AA chickens were randomly divided into 5 groups , which include normal group, OA model group, GYD low dose group, GYD high dose group, Finbid group, there were 8 chickens in each group, half of chickens were male. The chickens in GYD low dose group were fed decoction of GYD 5g/kg, those in high dose group were fed decoction of GYD 10/kg, those in Fenbid group were fed Fenbid 150mg/kg, those in normal group and OA model group were fed distilled water, all groups were fed twice. Detect activity of SOD and content of MAD from chicken serum in each group. Result: The activity of SOD from chicken serum in OA model group was significantly lower than in normal group(p<0.05).
