Qi Guard Tang Study The Impact Of Asthma In Mice Immune State

Asthma is a common chronic pulmonary disease which threat pubic health seriously. Inhaled corticosteroid is the most effect therapy for asthma by far, but it just emphasize local airway while ignore the whole body. Traditional Chinese Medicine(TCM) has the predominance in treating the human body as a whole and in the remission of a disease. It can regulate the disorder of the immunol system to alleviate the airway inflammation and prevent asthma through ''Fuzhengguben". Yiqihuwei Decotion is created by my advisor -Prefessor Hong Guangxiang. The theroy of the doction is based on Qiyangxuruo -the inner pathogeny of asthma. It is effective in the remission of asthma which is made up of Yupingfeng Decotion, Quizhi Decotion and Erxian Decotion. In this research we studied the influence of the disorder immunologic state of the local airway and the whole body.We selected the BALB / c mice as asthma model. The model is estabished via intraperitoneal injection and aerosol ovalbumin(OVA) for times to simulate the process of asthma. The cells in bronchoalveolar(BALF) and lung of model mice demonstrated that there was the infiltrate of inflammatory cells such as eosinophils(EOS), lymphocytes, macrophages and nutrocytes in ariway. So we have established the asthma model successfully.48 mice were devided into four groups at random: asthma model group, the doction group, corticosteroid group and normal group. The decotion group and corticosteroid group were treated from the first day. Collected the blood, BALF, lung and spleen 48 hours after challenged at the last time. CDS, CD4 and CDS T lymphocytes in peripheral blood were detected via flow cytometer(FCM).The level of serum soluble interleukin-2 receptor (sIL-2R), interleukin-4(IL-4) and interferon- Y (IFN- Y ) in BALF were detected via enzyme-linked immuosorbent assay(ELISA). IL-4mRNA and IFN- Y mRNA expression in spleen cells were detected via reverse transcription polymerse chain reaction (RT-PCR). The results were stated via the process of GLM in SAS6.12( a =0. 05).We discovered that : Yiqihuwei Decotion discreased the total number of cells and the proportion of EOS in BALF, alleviated the airway inflammation; decreased the proportion of CD4 T lymphocytes in peripheral blood and the ratio of CD4/CD8; decreased the level of serum sIL-2R to suppress the activation of CD4 T lymphocytes: deceased the level of IL-4 while creased the level of IFN- Y in BALF, accelerated the balance between Thl and Th2 subsets in airway; dereased the expression of IL-4mRNA while creased the expression of IFN- Y mRNA expression in splee cells, accelerated the balance between Thl and Th2 subsets in a whole.We conclude that Yiqihuwei Decotion can alleviate the airway inflammation, but also regulate the disorder of the immunol system and strengthen the alility of immunoregulation of the body. We suppose that Yiqihuwei Decotion maybe strengthen the effect of corticosteroid .
