Non-toxic goiter, such as single goiter and thyroid adenoma, not complicated with hyperthyroidism, is a common disease of endocrine system and often caused by psychic, environmental or genetic factors. Jiazhongfang is an experiential prescription of Professor Li Renxian for the treatment of goiter and shows good effect in clinic practice. In order to find out its modern mechanism of the prescription, in this research its clinic effectiveness was observed, and its effects on rat-model's thyroid function, microstructure and ultrastructure, thyrocytes apoptosis and proliferation, and protein express of relative genes of apoptosis were studied as well.Theoretic ResearchNon-toxic goiter belongs to the category of disease "Ying" of TCM. Single goiter belongs to "Qi Ying" and thyroid adenoma to "Rou Ying". Early in Qin and Han Dynasty, TCM already knew that Ying had a relation with water and soil in pathogeny and herbs containing iodine such as Haizao and Kunbu could be used for its treatment. In Tang and Song Dynasty, the ancestor knew that its pathogeny were associated with improper diet and disharmony of emotion, its pathogeneses were mainly Qi stagnation and phlegm obstruction and its therapeutic methods were soothing liver to activate qi and resolving phlegm to reduce swelling. In this period, Ying was divided into five kinds including stone, flesh, vein, blood and qi ying and thyroid of animal was creatively used. In Ming and Qing Dynasty, great many prescriptions for Ying were accumulated. Nowadays, many patients have to have their thyroid cut because of few medicines effective for this disease. But after operation, many of them have to take thyroid hormone for a very long time, even for lifetime, in order to maintain the normal function of thyroid and to prevent relapse. Thyroid hormone is also used to substitute operation, but it has not good therapeutic effect and easy to causehyperthyroidism. So to find out more effective and safe medicines is an important work. There are many reports of study on the treatment of this by TCM disease and some of them showed good therapeutic effect, but most of these reports did not have scientific design and so were weak in their conclusion.Clinical Research28 cases, completed clinical observation of 2 to 6 monthes period, were diagnosed as single goiter or thyroid adenoma (Qi Ying or Flesh Ying) and convinced by B-ultrasonic that thyroid anthorisma, or/and thyroid nodes existed, excluding hyperthyroidism and therioma. Based on the syndrome differentiation, cases of treatment group were divided into three types as the spleen deficiency and phlegm stagnation, the liver depression and qi obstruction and phlegm stagnation, and the others. They were treated with Jiazhongfang A, Jiazhongfang B, and combination of A and B respectively. 16 cases treated with L-T4 were selected as control. The result showed that in treatment group curative rate was 42.86%, effective rate 32.14% and total effective rate 89.29%, while in control group curative rate was 31.25%, effective rate 18.75% and total effective rate 62.50%. Effective rate of Jiazhongfang was superior to that of L-T4, indicating that the former is better than the latter in therapeutic effect. Jiazhongfang showed no side effect and its effect lasted longer than that of L-T4. The result also showed that Jiazhongfang was weaker than L-T4 in regulation of hormones such as T3% T4, FT3andTSH.Experimental ResearchThe purpose was to determine the effects of Jiazhongfang (combination of A and B) on each parameter of rat models. 18 rats in normal control group were perfused with tap-water 2.5 ml for 0 to 42 days and were killed at 0, 21st, 35th, 42nd day respectively. 56 rats were perfused with 1% 6-propyl-2-thiouracil (PTU) 1 ml for 7 to 42 days and 18 among them were killed at 14ths 21st day. 20 rats among the left were taken out as treatment control group from 21st day, perfused with 2% PTU 1.5 ml and Jiazhongfang liquid 1.5ml for another 21 days (22nd to 42nd) and killed at 28th, 35th and 42...
