The Experimental Study Of Andrographolide Anti-respiratory Virus In Vitro And Anti-influenza A Virus In Vivo

Especially respiratory virus often product variation and increase pathogenicity, or lead new diseases. For example, in recent years, SARS, human avain influenza, influenza A virus have damaged people's health and lifes. While it even effect stability of social and national economy. SO many people pay attationto it.This paper begins at the structure of four respiratory virus. It described the replication of four virus, epidemiolog features, Pathogenic mechanism, clinical manifestation, diagnosis and the research progress of influenza vaccine. On research of pharmaceutical anti- respiratory virus, this paper described different mechanisms and drugs between traditional chinese medicine and western medicine. And furthermore showed that traditional chinese medicine has great advantages.Experiment StudyObject:To investigate the effect of Andrographolide anti-respiratory virus in vivo and vitro, also anti-influenza A virus in vivoMethod:In vitro, at first, study the influences that Andrographolide inhibitory influenza A Virus by culture MDCK. The second, study the influences that Andrographolide inhibitory Rhinovims and Adenovirus by culture HEL cells. The third, study the influences that Andrographolide inhibitory respiratory syscytial virus by culture A549 cells. In vivo,we observed the effect of the mice lung index, the death rate of mice, TNF-a,αIL-10, Neuraminidase and Nucleic acid content what the Andrographolide do.We use influenza A virus drop in the mice nasal cavity.Result:1,Experiments in vitro:At the first, we use egg inoculation amplificate virus and detect the virulence by blood agglutination experiment and plaque forming assay. According to the result of blood agglutination experiment, we calculate the TCID50 is 3.67. In order to decide Andrographolide's maximum of no toxicity concentration,we divided it into different concentration to test. And then observed the preventive effect, protective effect,direct cytotoxic effect and the dynamic of prevent virus replicate to research the effect of Andrographolide anti-influenza A virus. The result showed: Andrographolide has the effects at concentration of 125g/L and reduced two virus agglutination titer. Andrographolide inhibit Rhinovims (TCID5=5.5) and Adenovirus (TCID5= 5.67) at concentration of12.5 g/L and 6.25 g/L. The therapeutic index are 17.99 and 46.89 Andrographolide inhibit respiratory syscytial virus (TCID5=5.0) at concentration of 3.12g/L.2,Experiments in vivo:Andrographolide at the high dose of 15g/kg can reduced the mice lung index, alleviated pulmonary consolidation, improved the symptoms that caused of viral pneumonia, raised the mice survival rate, reduced the content of TNF-αand improved the content of IL-10. The result showed that this drug can raise the mice's phylactic power and improve immune dysfunction that caused of virus infection. It can not reduced the content of neuraminidase (NA) and showed the effect target was not NA. The result of fluorescence Polymerase Chain Reaction(PCR) indicated that Yuye detoxifi- cation particle can inhibit the mice lung tissue virus that infected from influenza A virus. According to this inhibitory effection,it can alleviate mice body injury.ConclusionThe results of this experimental research demonstrated that Andrographolide has better inhibitory effection in anti-influenza A virus research at concentration of 125g/L in vitro. And it showed different kinds of anti-virus effections.It can inhibit Rhinovims and Adenovirus at concentration of 12.5 g/L and 6.25g/L.Also inhibit respiratory syscytial virus at concentration of 3.12g/L. While in vivo experiments, influenza A virus can be suppressed at the high dose of 15g/kg. At this concentration, it reduced the symptoms of viral pneumonia, prolong the mice's life. In a word, Andrographolide not only has good therapeutic effects in vitro,but also is an effective traditional chinese medicine anti-influenza virus in vivo and provide reliable laboratory basis for the clinical widely applied.But the effection of Andrographolide anti the other respiratory virus in vivo have to be proved.
