Objective: To discuss the protective mechanism of acanthopanax on learning and memory in sleep deprivation rats from the synaptic plasticity and cholinergic neurotransmitter point of view.Methods: Take rats as object of study, to investigate the dose-response relationship and time-concentration of Acanthopanax increasing learning and memory in normal rats by the hexagonal maze method. Based on this, established the model of rapid eye movement sleep (REMS) deprivation in rats by using improved multi-platform, and Acanthopanax was given. Ethology and nerve electrophysiology methods were performed to study effects of Acanthopanax on the synaptic plasticity and neurotransmitter in sleep deprived rats.Results: (1)The highest recognition rate appeared on 7days'administrating Acanthopanax of 5.6g/kg continuously on normal rats. (2)After 96h REM sleep deprivation, the recognition rate of model rats decreased, Acanthopanax increased the recognition rate.(3)After 96h REM sleep deprivation, long-term potentiation (LTP) of model group rats was suppressed, but Acanthopanax can enhance neuronal plasticity in hippocampus of sleep deprivation rats.(4)After 96h REM sleep deprivation, AChE activity of the model group in the hippocampus and cortex increased, but Acanthopanax can decrease AchE activity in rats of sleep deprivation.(5)After 96h REM sleep deprivation, 5-HT and 5-HIAA level in hippocampus and cortex of model rats increase, but NE in hippocampus and cortex, DA in cortical decreased. Acanthopanax decreased the level of 5-HT, and increased NE, 5-HIAA, 5-HIAA/5-HT in hippocampus and cortex, and DA in cortical.Conclusion: Administrating Acanthopanax can increase the ability of learning and memory in normal rats, and the effect appears on the third day,and the best case is 5.6g/kg for 7days'continuously administrated. Acanthopanax can effectively fight against learning and memory decreased in rats of REM sleep deprivation, by means of enhancing synaptic plasticity and regulating disordered central monoamine neurotransmitters.
