A gorgonian known as sea whip or sea fan, is a sessile animal of the Subclass Alcyonaria, Class Anthozoa, Phylum Coelenterate found throughout the oceans of the world, especially in the tropics and subtropics from inter-tidal to as deep as 4,000 meters. There are 45 species from seven families of gorgonians in China, mainly located in Hainan, Guangdong, Zhejiang and Taiwan coast. Studies in the past half century showed that a variety of novel and bioactive chemical constituents were gained from the gorgonian. The gorgonian Muricella sibogae were collected from the coast of Weizhou Island, Guangxi province, China in April,2006. This paper systematically investigated the chemical constituents and biological activities of the gorgonian Muricella sibogae.Isolation and structure determination:Monitoring by bioassay-guided isolation, the MeOH extract of the gorgonian Muricella sibogae was chromatographied on silica gel, Sephadex LH-20 and semi-preparative HPLC respectively, then forty seven compounds were isolated and purified from the gorgonian Muricella sibogae. These compounds were identified on the basis of spectral methods (NMR, MS, IR, etc.) and their physicochemical properties. Among them, there were six new compounds 24-ethyl-calicoferol E(1),24-methyl-calicoferol E(2),28-methyl-Calicoferol D(3), 6-methoxy-calicophirin C(4),6-eacetoxyl-7-aldehyde-calicophirin C(5),6-eacetoxyl-calicophirin C(6). And the known compounds were structurally elucidated as: caffeine(7), cholesterol(8),24-melthyl-cholest-5,22-dien-3β-ol(9),Cholest-5,22-dien-3β-ol(10),ergosta-5,24(28)-dien-3β-ol(11),(Z)-Stigmasta-5,24(28)-dien-3β-ol(12),stigmast-4-en-3-one(13),stigmasta-4,22-dien-3-one(14),(3-sitosterol(15), stigmasterol(16),7-oxo-(3-sitosterol(17),3β-hydroxy-stigmast-5,22-dien-7-one(18), ergosterol proxide(19), stigmast-4-ene-3,6-dione(20), Ergosta-4,24(28)-diene-3-one (21), stigmast-4-ene-3,6-dione(22),24-norcholesta-4,22-dien-3-one(23), dendronesterones C (24),Ergosta-1,4,22 -trien-3-one(25), Ergosta-1,4-dien-3-one (26), calicoferol A(27), Calicoferol D(28), calicoferol E(29),24(28)-en-calicoferol E (30),24(28)-en-29-Me-calicoferol E(31), 13-deacetoxyl calicophirin B(32), Ophirin (33), calicophirin C(34), benzoic acid(35), 4-Hydroxy-benzoic acid(36), methyl-Phydro-benzoate(37), batyl alcohol(38), uracil(39), thymine(40),2'-deoxyuridine(41),2'-deoxythymidine(42),2'-deoxyadeno-sine(43), Adenine(44), dibutyl phthalate(45), diisobutyl phthalate(46),1,2-benzenedicar boxylic acid bis (2-ethylhexyl) ester(47).Biological activities:The methanol extract of the gorgonian Muricella sibogae was evaluated for the antimicrobial activity against the strains of vibrio anguillarum. The results show that the fractions of BL-4, BL-5, BL-6 were the main parts of the methanol extract for the antimicrobial activity. The methanol extract of the gorgonian Muricella sibogae was valuated for the cytotoxicities against P388 and BEL-7402 cancer cell lines by the MTT method. The results show that the fractions of BL-2, BL-3, BL-4, BL-9, BL-10 were the main parts of the methanol extract for the cytotoxicities activity. The compounds gained from the gorgonian Muricella sibogae are in the detection of biological activity.
