Dendrobium officinale is a traditional valuable Chinese herbal medicine, its natural resources will be dried up, and it has been listed as one of protected plants which danger for extinction. Investigation on its germplasm resources will be advantageous to introduction of artificial planting with purpose. Induction of mutation with physics and chemical methods are the common use methods for Chinese herbal medicine species improvement, Researches on these aspects will be advantageous to sustainable growth on the D. officinale industry. The germplasm resources of D. officinale can not get away from its micropropagation, and micropropagation in industrial way is the foundation for planting in large scale to turn, how to lower the production cost is a foundation stone that maintains the D. officinale industry to develop healthily. Because of the long cultural period of D. officinale, how to resolve effectively the contamination is an important aspect to decline the cost.Analysis on biological characters and active ingredients of 8 varieties of D. officinale in different regionsWe compare the height , stem thickness, leaf length , leaf width , leaf area, and ratio of leaf length and width among 8 varieties of D. officinale in different regions , and also measure the water content, total polysaccharide, water-soluble polysaccharide and cellulose in D. officinale mentioned above.Conclusion: There are many differences among 8 varieties of D. officinale in biological characters;D. officinale in Yandang have good agricultural characters and high level of active ingredients and it is suitable for planting in Zhejiang. Inducement of tetraploid D. officinaleProtocorm of D. officinale were directly soaked in colchicines solution with different concentrations and different treated time. Analysis on the effects of colchicines were checked, ploid analysis carried by flow cytometry and root tip chromosome check.Conclusion: The tetraploid plants were obtained .The optimum concentration ofcolchicines is 0.05% with 10d treated time.The effect of 137Cs γ-ray irradiation on the growth and regeneration of D. officinaleprotocorm.The irradiation effects which include LD50 .growth performance, multiplication rateand variation rate with doses (0,25, 50, 75, 100Gy) on D. officinale protocorm werechecked.Conclusion:LD50 is about 70Gy. The larger the dose irradiated, the more depressed theplant growth. The major reason of variation is change in DNA level.Research on optimizing micropropagation craft of D. officinaleBy the innovation on cultural container, we introduced pile line operation onmicropropagation, and also we invent the multi-function frame and adopt the natural lightfor the culture.Conclusion: By adopting the new cultural container and new craft, the product efficiencywas raised more than 300%;and the energy consume was lowered 50 times than traditionalway.Prevention and cure of isothiazolone to bacterial contamination in commercialmicropropagation of D. officinaleBacterial contamination is a serious problem in commercial micropropagation. The result shows 50 mg-L-1 isothiazolone can kill microbe when put it on the surface of media, and promote growth of the test tube seedling(up 3~3. 5cm) under this concentration, such as increase the fresh weight, roofnumber and stem thick., But it restrict the root growth longer strongly.
