This dissertation consists of six chapters focusing on the study of extraction method, physical and chemical characters and biological activities of forsythiaside, which is the effective ingredient of Forsythia suspensa(Thunb)Vahl.. The contents are as follows: 1. The research developments of Forsythia suspensa(Thunb)Vahl., inhibitors of elastase and natural antioxidants were reviewed. 2. Forsythiaside has been obtained by using Macro-reticular resin D101 and low pressure liquid chromatography,which have some obvious advantages:saving solves,time and having high performance. 3. Physical and chemical characters of forsythiaside were studied by using UV and fluorescence spectrum in different conditions. The results shown that forsythiaside is very unstable under the condition of alkali(pH=9.40), and relatively stable when pH is 6.86 and temperature is about 60℃. When temperature is above 80℃and pH is above 6.86, forsythiaside shown very unstable. But it shown relatively stable when temperature is 80℃and pH is 4.03. 4. Extraction technology of forsythiaside was detailed studied. There are many advantages using this extraction technology: (1) the used solvents are safety and innoxiously, and do not contaminated environment. (2) Macro-reticular resin can be used many times and regenerated easily. (3)saving solves,time and having high performance, and content of forsythiaside was over 40 percent. 5. Using the method of agarose diffusion, the inhibition of Porcine Pancreatic elastase by Chinese herbs and forsythiaside, chlorogenic acid and other effective ingredients of Chinese herbs has been studied. The results shown that forsythiaside, chlorogenic acid and other effective ingredients of Chinese herbs could inhibit the activity of Porcine Pancreatic elastase, and the active matter of radix scutellariae extracted by acetone can inhibit the activity of elastase very strongly. Further research discovered that active matters of radix scutellariae extracted by acetone can not ameliorate emphysema symptoms of experimental big rat, and the herbal medicine composed by many herbs ameliorate emphysema symptoms of experimental big rat better than forsythiaside. 6. The scavenging effects of the forsythiaside and its analogs on HO·,H2O2 and O-2·were studied by using chemiluminescent and NBT methods. The results show that scavenging effect of them on activated oxygen all is strong. The relationships of structure –activity studied with quantum chemistry (PM3, RHF) indicate that the reactivity of phenolic hydroxyl, which in ring A and B of forsythiaside, is almost identical. Further research discovered that forsythiaside can eliminate free radical in the system of lard, and can prolong storage term, and scavenging effect of forsythiaside on activated oxygen is better than BHA appreciably. 7. The extraction of alkaloids from Sophora flavescens Ait. with CO2-SFE was studied. The results showed that the optimum conditions of CO2-SFE were: pressure 30MPa, temperature 45℃, time 5h for the extract, with taking NH4OH as alkalizer and MeOH as entrainer, and the extraction rate of supercritical CO2 fluid extraction was as about 2.4 times as that of traditional solvent extraction.
