All organisms respond to heat and a wide variety of other stresses by inducing the synthesis of a group of proteins called the heat- shock proteins or hsps. This response is universal, existing in every organism in which it has been sought, from archaebacteria to eubacteria, from plants to animals. Hsps can be divided into several families according to conserved structures and functions: hsp110, hsp90, hsp70, small hsps and ubiquitins. The regulation of hsps is mainly on the transcription level. Now, the study of hsp genes' transcription is a hot issue. Because the heat shock gene regulatory circuit has several features that make it an attractive model for investigating the activation of transcription, at least some of the principles identified in the study of transcription of heat shock genes are likely to be of general implication to transcription regulation. Comprehensive studies from many labs provide a strong foundation of information on the structure of these genes, the DNA sequence elements and protein factors that participate in the regulation of their transcription, but the studies concentrate on the hsp70 gene.Hsp90 family is one of the most important families. The 80-90kD class of hsps have been recognized as cytoplastic proteins that are abundant in the absense of stress and induced to higher levels of synthesis by heat shock (hs). They have been found in association with some regulatory and structural proteins in avian and mammalian cells and tissues. Therefore, the study of hsp90 will be of clinical and theoritical implications. Two forms of hsp90 that differ slightly in molecular mass have been identified in murine and human, hsp90α and hsp90 β, the mRNA of which are referred to 2.95kb and2.7kb. Hsp90α contains 732 amino acid residues with a predicted unmodified...
