The isolation-fusion technology of protoplasts of microbial cell is a kind of developing technology growing on the basis of the cell fusion with gene technology. In the dissertation some breaking research had been trying to do in this area. Two of the yeast were chosen to study the isolation-fusion technology of protoplasts, One was Saccharomyces cerevisiae which was able to grow in a highotemperature (42 C) and rich-alcohol (15%) surrounding, and another was Nectaromyces sp. which was able to resist high osmotic pressure (55% glucose). We tried for creating a new way of genetic breeding and providing a method for industrial microbial breeding, as well as setting up an experimental model of transfering isolated nuclei and anucleated system of eukaryotic organisms, to supply a new sort of technology for researching distant hybridization and genetic recombination on the level of molecule or cell.The main contents of the dissertation are expounded as follow:1.The effects of several factors, such as cell growth phase, lytic enzymes, concentration of enzyme, time and temperature of enzyme treatment, osmotic stabilizers, osmotic pressure change, pH of buffer, growth media, keeping activity of protoplasts, precursor of yeast cell wall syntheses, modes of incubation etc., were investigated on the formation and regeneration of protoplasts. The optimum conditions preparing highly active protoplasts of S. cerevisiae were established.
