Objects: To indicate the differences of action spectra between traditional formulas and modern formulas by comparing the differences of effect between the active frictions of Ge Gen Qin Lian Decoction and the original formula on experimental metabolic syndrome. To offer experimental evidences and research considerations for new medicine suited for modern diseases spectra among traditional formulas as well as new pattern for the second development of classical formulas.Methods: To establish metabolic syndrome model of rats by giving 10% of diabetin solution. To establish models and index system of medicine effect evaluation according to the diagnoses standards of metabolic syndrome. To test the sensibility of insulin, blood glucose, fat of the internal organs and blood fat, etc. of metabolic syndrome model rats by the pharmacological , biochemical, histological and molecule pathological ways. Then to evaluate the effect of medicine, and to compare the action spectra and the effect intensity of all the components of MS between Ge Gen Qin Lian Decoction and its active frictions. To analyze the position of action and mechanism of them and to explore the possible reasons of the differences of action spectra between Ge Gen Qin Lian Decoction and its active frictions by testing tumor necrosis factor in the blood serum, hydrocortisone, C peptide, hormone, leptin, and insulin receptor of hepatic cell, etc. of the MS rats.Results: After giving the rats 10% of diabetin solution for six weeks, the weight as well as blood glucose and triglyceride of the model group rats increased dramatically, with the decreasing of the sensibility of insulin index, which comply with the MS diagnostic indications. The high, middle and low dosages groups of the active frictions of Ge Gen Qin Lian Decoction can increase the glucose tolerance and glucose infusion rate significantly and reduce the level of blood glucose, serum insulin and leptin of the model group rats. The middle and low dosages groups of the active frictions of Ge Gen Qin Lian Decoction can increase the level of high density lipoprotein obviously, decrease the lever of total cholesterol, FFA, TNF-α, C-P and ACTH as well as the weight of fat in the abdominal cavity, and increase the average gray scale of agentic goals of the insulin receptor of hepatic cellular. The low dosages group can reduce the triglyceride observably. The middle dosages group can reduce the weights markedly. The high dosages group can reduce the low density lipoprotein dramatically. The high, middle, low dosages groups of the active frictions of Ge Gen Qin Lian Decoction and the Ge Gen Qin Lian Decoction group can dramatically decrease the ratio of fat in abdominal cavity and weights , at the same time the concentration of the serum hydrocortisone. The effect of the middle group is better than that of the Ge Gen Qin Lian Decoction group. The middle dosages group of the active frictions of Ge Gen Qin Lian Decoction can observably improve the apomorphosis of the liver fat.Conclusions: The MS model can be successfully established when giving the rats 10% of diabetin solution for six weeks. The active frictions of Ge Gen Qin Lian Decoction can increase the sensibility of insulin of MS rats, improve the lipid metabolism disorders, reduce blood glucose, lessen the abdominal fatness and control the main components of MS effectively. These effects are related to the roles of increasing the density of insulin receptor, improving the leptin resistance and reducing the anti-insulin hormone , which can improve the pre-receptor and receptor deficiency of insulin. The same dosages of Ge Gen Qin Lian Decoction has no apparent effect to other indexes expect that it can reduce the ratio of fat in abdominal cavity and weights. The pharmacological action spectra are different obviously between the Ge Gen Qin Lian Decoction and its active frictions, due to the different preparations. The reason may be the matching relationships of active components are changing resulting from the interactions of drugs.
