PARTâ… Analysis of the inhibitory effect in vitro of anti-HP Lactobacilli acidophilus on HP isolated from different chronic gastropathy[Objective] To prove Lactobacilli acidophilus obtained from our previously study and can inhibit international standard strand H PSS1 and NCTC11637 ,still have inhibitive effect to HP clinical isolates obtained from different chronic gastritis patients.[Method] We cultivated anti-HP Lactobacilli acidophilus of different density in solid and liquid mediums together with HP obtained from 15 different chronic gastropathy .The diameter of inhibitory rings ,HP colony count and the change of the activity of urease indicated the inhibitive effect of anti-HP Lactobacilli acidophilus to HP clinical isolates. At the same time, we tested the HP virulence factors to observe the relation between inhibitive effect of anti-HP Lactobacilli acidophilus and HP virulence factors.[Result] Both anti-HP Lactobacilli acidophilus have general inhibitive effect to HP clinical isolates from different gastritis sources. Furthermore, as the density of anti-HP Lactobacilli acidophilus, the diameter of inhibitory rings increased gradually, HP colony count decreased obviously, activity of urease cut down gradually, and were significantly different from control group, P<0.05.[Conclusion] Lactobacilli acidophilus which were obtained from our previously study have obviously inhibitive effect to HP isolated from different gastropathy. This inhibitive effect does not have relation with the kinds of chronic gastropathy and HP virulence factors.PARTâ…¡Analysis of inhibitory effect of anti-HP lactobacilli acidophilus on the adherence function to SGC-7901 cells and IL-8 production of HP isolated from different chronic gastropathy[Objective] To examine the adhesive ability of anti-HP lactobacilli acidophilus to the gastric epithelial cells and its antagonism to Helicobacter pylori of chronic gastropathy. Investigate the antagonism of anti-HP lactobacilli acidophilus to IL-8 secrets introduced by Helicobacter pylori. [Methods] Microscope and Scanning electron microscope were used by analyze the adherence index of the different concentration anti-HP lactobacilli to the gastric epithelial cells and as well as the changes of the adherence index of the epithelial cells after antagonism with the anti-HP lactobacilli acidophilus, the vitality of epithelial cells after the adherence of anti-HP lactobacilli acidophilus was observed by trypan blue assay. The ELISA technique was also used to study the inhibitory effect of the anti-HP lactobacilli acidophilus from the IL-8 secrets by epithelial cells.[Result] Anti-HP lactobacilli acidophilus had adhesive ability to gastric epithelial cells and the vitality of epithelial cells did not decrease. Anti-HP lactobacilli acidophilus had antagonistic ability to Helicobacter pylori and had inhibitory effect to IL-8 secreting introduced by Helicobacter pylori.[Conclusion] Anti-HP lactobacilli acidophilus had adhesive ability to the gastric epithelial cells and had antagonistic ability to Helicobacter pylori meanwhile inhibited IL-8 secreting by Helicobacter pylori.PARTâ…¢The prevention and therapeutic effects of anti-HP Lactobacilli acidophilus on C57BL/6 mice with HP infection[Objective] To examine the protective and therapeutic effects of anti-HP Lactobacilli acidophilus on HP SS1 infection in vivo . [Methods] One group of C57BL/6 mouse was given anti-HP Lactobacilli acidophilus first and then HP. The other group was just contrary to the above. Observe the activity of urease, the HP colonization , the infiltration degree of mononuclear and lymphocytes , and the separating culture result of HP in the gastric mucosa of the mouse. Investigate the infection rate of HP and the local traumatic degree, confirm that anti-HP Lactobacilli acidophilus has the effect of prevention and treatment on HP infection.[Result] The group which was given anti-HP Lactobacilli acidophilus beforehand , could decrease the HP infection rate of the C57BL/6 mouse obviously. It has significant difference in comparison with the HP infection only group (P<0.05) , and it could also reduce the traumatic degree of the mouse infected by HP in the gastric mucosa . After treating with anti-HP Lactobacilli acidophilus, the HP infected group of mouse showed a low HP colonization density and local traumatic degree in the gastric mucosa, with significant difference compared with the control group (P<0.05) . The serum HP-IgG antibody titer decreased , and when treated for 9 months it became the most ,with significant difference compared with the control group (P<0.05) . The group given anti-HP Lactobacilli acidophilus after HP eradication therapy showed a low reinfective rate with significant difference compared with the control group (P<0.05).[Conclusion] Prophylactic use of anti-HP Lactobacilli acidophilus can significantly decrease the HP infection and reinfection of C57BL/6 mice and can also decrease the degree of inflammatory reaction after HP infection in the gastric mucosa. To the HP infected mouse, anti-HP Lactobacilli acidophilus can decrease the degree of inflammatory reaction and the serum HP-IgG antibody titer.
