Purpose : To observe the effect of eye acupuncture therapy on the expression of neurotrophic factors such as BDNF,NGF,GDNF,bFGF in the brain of CI/RI model rats,to explore the mechanism of eye acupuncture in improving ischemic stroke,and to provide evidence support for the wider application of eye acupuncture in clinic.Material and method:This experiment selected 72 adult SPF grade SD rats,which are36 male and 36 female rats,respectively.After 7 days of adaptive feeding,all animals were randomly divided into 2 groups according to 1:3 ratio :18 sham-operated groups,The model group was 54.A CI/RI model was prepared by modified thread suppository method in model rats.The sham-operated rats were treated with the same procedure but no substantial embolism.After the model preparation,two groups of rats were judged by Zea Longa5 scoring criteria.Each random sample 1 used TTC staining method to determine the presence or absence of infarct focus.After the model was established,the model rats were divided into model control group,non-menstrual group and eye acupuncture group by random digital table method.The sham operation group and model control group were not treated,normal feeding until the end of other groups of intervention.Ophthalmic acupuncture treatment group was treated 6 hours after reperfusion,12 hours after reperfusion and 24 hours after reperfusion,and a uniform reinforcing-reducing was applied.Acupuncture was performed in the non-acupoint group about 5 mm outside the acupuncture point area.The intervention time was consistent with the eye needle group.After the intervention,the general conditions of rats in each group were observed and behavioral evaluation and index determination were carried out.Conduct behavioral testing by Zea Longa5 scoring method;HE staining was used to observe the pathological changes of rat brain tissue.The positive expression of ischemic penumbra BDNF,NGF,GDNF and bFGF in rats was detected by immunohistochemistry.ELISA determination of ischemic penumbra BDNF,NGF,GDNF and bFGF protein in rats.Nissler staining was used to observe the damage of neurons,and count the positive neurons.Results:1.After modeling,the neurological deficit score showed that the model group score was significantly higher(P < 0.01)than the sham operation group,and the difference was statistically significant.2.After modeling,the results of TTC staining showed that compared with the sham operation group,white mass appeared in the brain tissue of the model group,indicating the existence of infarct focus.3.The behavioral score after intervention showed that the score of eye acupuncture treatment group was significantly lower than that of model group(P < 0.01),and there was no significant difference between non-menstrual and non-acupoint group(P>0.05).4.After intervention,the results of HE staining showed that there was no cell edema and degeneration in the sham operation group;the cell integrity of the model control group was destroyed,which was in sharp contrast with the sham operation group.The neuro of nerve cells in the eye needle group was clear and the recovery trend was higher than that in the model group.5.The results of immunohistochemistry showed that the expression levels of ischemic penumbra BDNF,NGF,GDNF and bFGF in the model control group decreased significantly(P<0.01).There was no significant difference in BDNF?NGF?GDNF and bFGF expression between non-acupoints group and model control group(P>0.05).Compared with the model group,the expression of BDNF,NGF,GDNF and bFGF in the eye acupuncture group was significantly increased(P<0.01).6.The results of ELISA method showed that the contents of ischemic penumbra BDNF,NGF,GDNF and bFGF in model control group were significantly decreased(P<0.01)There was no significant difference in BDNF?NGF?GDNF and bFGF expression between non-acupoints group and model control group(P>0.05).Compared with the model group,the expression of BDNF,NGF,GDNF and bFGF in the eye acupuncture group was significantly increased(P<0.01).7.The results of Nissler staining showed that the number of Nissler bodies in the sham operation group decreased sharply and the number of nerve cells was lost(P < 0.01).Compared with the model control group,there was no difference between the non-acupoints group and the model control group(P>0.05).Compared with the model control group,the number of Nissler bodies increased and the number of nerve cell positive increased significantly(P<0.01).Conclusion:1.Eye acupuncture therapy can effectively improve limb dysfunction in CI/RI model rats and alleviate brain tissue injury to some extent.2.Eye acupuncture intervention liver area,kidney area,upper energizer area,lower energizer area can obviously up the expression level of neurotrophic factors such as BDNF?NGF?GDNF?bFGF in cerebral ischemic penumbra area of CI/RI model rats.3.Eye acupuncture therapy can effectively improve the neuronal function and increase the number of positive neurons in the ischemic penumbra area of CI/RI model rats,which may be related to the neurotrophic factor providing nutrition for neurons and promoting their proliferation.Induction of angiogenesis to reduce secondary injury of neurons,affecting neuronal outcome.
