Objective To observe the effect of transplantation of the marrow stroma cells (MSCs) modified with gene of NT4-GFP into hippocampus of AD rats. Methods AD models were made by transecting bilateral fimbria-fornix ; The marrow stroma cells were cultured and purified in vitro.; NT-4 geng was constructed and cloned into the marrow stroma cells(MSCs); the MSCs MSCs modified with-NT-4 were separately transplanted into the hippocampus of AD rats; the behavior tests and auditory evoked potential P50 were carried out; Number of ChAT positive neurons were counted and Gene products of NT-4 were tested as well as the survival of MSCs was observed.Results AD model were established successfully Auditory evoked potential P50 was changed significantly; MSCs-NT-4 can stably express NT-4 both vitro and vivo . Improvement of Behavior and neuroelectrophysiology function of MSCs-NT-4 transplantation is more obvious than that of MSCs transplantation. Conclusions MSCs are donator of transplantation for AD, Transplanting MSCs-NT-4 into hippocampus of AD rats can obviously improvement cognition function and neuroelectrophysiology function.
