Objectives1. To establish the procedures for determination of free L-carnitine(FC), acetyl-L-carnitine(AC) and, total carnitine(TC) in human plasma and urine by high performance liquid chromatography(HPLC).2. To determine the level of FC, AC and, TC in plasma and the urine excretionamount of FC, AC and, TC in 24 hours of 20 healthy volunteers.3. Multi-center survey on the changes of FC, AC and, TC level in plasma and urineexcretion amount of FC, AC and, TC in 24 hours of surgery-stressed patients (with 120 cases).Methods1. Pre-column derivatization was performed with 1-aminoanthracene in the presenceof 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide hydrochloride as catalyst after esterified carnitine being hydrolyzed into free carnitine. C18 column was for plasma determination and positive ion exchange column for urine.2. To determine FC and AC in plasma, TC in plasma, FC and AC in urine and, TC inurine respectively by HPLC.3. Twenty-four hours urine was collected and fast blood sample was drawn in threesuccessively days from 20 volunteers with standard diet in the same time.4.
