Objective:To investigate the protective effects of a Chinese Medicine formula "Xianzhen Tablet Composita" (XZTC) on the kidney in streptozotocin-induced diabetic rats. The experiments include two parts. Part one: Effects of XZTC on blood glucose, glycosylated hemoglobin (HbA_k), renal function and pathologic picture of streptozotocin (STZ)—induced diabetic rat Part two: Effects of XZTC on advanced glycation end products (AGEs) and mRNA expression of AGE-specific cellar receptor (RAGE) in renal tissue of STZ -induced diabetic rats. Methods:1. Experimental diabetic conditions were induced in male Wistar rats by intraperitoneal streptozotocin in a single dose of 60mg per/kg body weight. The rats with blood glucose level above 16.7mmol/L were chosen as diabetic rats.2. In part one, the diabetic rats were randomized to the following four groups, untreated diabetic group, treated diabetic group with west medicine (Benalapril plus Gliquidone, positive control group), treated diabetic group with high dose XZTC (XZTC high dose group), and treated diabetic group with low dose XZTC (XZTC low dose group). Another group of nondiabetic rats matched for age and body weight with the diabetic rats was as normal control group. In the 12 th and 24 th week of diabetic state, blood glucose, HbA_k, Alb, Scr and BUN, kidney index (kidney weight /body weight) of each group were checked, pancreatic and renal pathologic changes were observed under light or electron microscopy. In part two, the rats were randomized to the following four groups: normal control group, untreated diabetic group, treated diabetic group with XZTC (XZTC group), and treated diabetic group with aminoguanidine (AG group, 100mg/kg body weight per day). In the 12 th week of diabetic state, the amount of AGEs and the level of mRNA expression of RAGE in renal cortex of each group were investigated respectively by fluorescent measurement, reverse transcription polymerase chain reaction (PCR).Results:1.
