In order to explore the relation between the CD44V gene abnormal expression and splicing and the clinicopathologic feature, the expression of CD44V of normal colorectal mucosa, adenomatous polys (precancerous disease) and colorectal carcinoma was detected using the technique of reverse transcriptase followed by amplification by PCR and cDNA hybridization. On the other hand, immunohistochemical method was used to detect P53 protein and PCNA in formalin-fixed, paraffin-embedded tissue specimens of colorectal carcinoma, and serologic assay was used to measure CA242 and CEA in the serum of the patients with the cancer. The clinical significance of CD44V, P53, PCNA, CA242 and CEA was discussed in early diagnosis and estimation of metastasis of the colorectal carcinoma. The results showed that CD44V mRNA was expressed in 65 of 67 (97%) primary cancers, in all metastatic tumors (20/20), in 5 of 7 (71. 4%) precancerous disease, and was not expressed in 15 cases of normal mucosa. An increased number of more intensely labelled, larger molecular weight bands in 10 of 20(50%) primary cancers, in 18 of 20 (90%) metastatic tumors were showed in all cancer tissue specimens. In...
