Objective: To investigate JianWeiYuYang granule(JWYY)'s effects on peptic ulcer recurrence through clinical research and animal experiment, meanwhile inflammation and the expression of NF-κBp65, β-defensin-2 in gastric mucosa were determined to reveal the mechanisms of JWYY's effects on ulcer recurrence from the view point of inflammation.Method:1. Clinical research: 50 Hp-positive PU patients were confirmed by endoscopy, histological inspection and urease test , and the differentiation of symptoms and signs of the traditional Chinese medicine is the stagnation of liver-QI with deficiency of the spleen type. These patients were divided randomly into JWYY group and western medicine group, 25 patients each group. 17 Hp-negative PU patients were set as Hp-negative PU group. Another 8 inspectors whose gastric mucosa was roughly normal without Hp infection were set as normal group. Patients of JWYY group were treated by JWYY 3g, 3 times per day , totally 6 weeks. Patients of western medicine group was treated by amoxicillin capsules 1.0g, 2 times per day; metronidazole 0.4g, 2 times per day, both for 7 days; omeprazole 20mg, 2 times per day, totally 6 weeks. Gastroscope was rechecked 4 weeks later after drug withdrawal, curative effect under gastroscope,eradicative rate of Hp,clinical curative effect in Chinese medicine syndrome and quality of life were evaluated. Whenever gastroscope was undertaken, some gastric mucosa at the site of sinus ventriculi were taken out for Hp urase test, HE staining, Warthin-Starry silver staining , detection of NF-κBp65mRNA, β-defensin-2mRNA and protein by in situ hybridization , immunohistochemistry and Western blot.
