Experimental Study Of Efficacy And Mechanism Of RJSJ On Anti-hepatoma

In this paper the experimental study on the anti-hepatoma efficacy and mechanism of Ran Jian San Jie (softening and resolving hard mass) was reported.1. Ran Jian San Jie prescription of Chinese traditional medi-cine(RJSJ-PM) composed of Carapax trionycis, Pseudobulbus cremastrae seu pleiones. Herba scutelarae barbatae(?) Rhizoma curcumaeetc. By using water-extraction and ethanol-precipitation,the 6%extractive of crudedrugs wasprepared. Dose tolerance in mice was30g/kg for oraland 8g/kg for intraperitoneal medication.2. RJSJ-PM had some inhibitory effect on the experimental hepatocarcinoma.(1) In vitro,for human hepatoma BEL-7402 cells ED_? of RJSJ-PM was38.5ug/ml for inhibiting the colony formation and 54.0ug/ml for counting the survival cells. RJSJ-PM (<75ug/ml) combined with 5-Fu (<5uM/ml) showed a coordinative inhibitory effect (CDI≤0.85, p<0.01).(2) With different dosage of RJSJ-PM,1.8g/kg.d,3.6g/kg.d and 7.2g/kg.d, mean inhibiting growth rate of tumor (HepA) in BALB/C mice wasl6.6%(p>0.05), 32.0%(p<0.05) and 49.8%(p<0.01), and the mean ratio(T/C) of delaying survival time of the mice bearing tumor (HepA) was1.09 (p>0.05), 1.35 (p<0.05) and 1.59 (p<0.01), respectively. The mean ratio(T/C) of delaying survival time of the mice bearing ascites(HepA) with 3.6g/kg.d and 7.2g/kg.d was 1.10 (p>0.05) and 1.35(p<0.05), respectively.3. The mechanism of RJSJ for antihepatocarcinoma was studied in two aspects as follows:(1) Inhibition of hepatoma cell growth:By optical and electron microscope investigation, a lot of vacuolar degenerated tumor cells (HepA) appeared in RJSJ-PM group (7.2g/kg.d). Besides this, also appeared a lot of expanded endo-plasmic reticulum and mitochondrion, autophagic vacuoles, concentrated broken chromosome near by the nuclear membrane, which suggested that the apoptosis of the HepA cells might be induced by RJSJ-PM.By flow cytometric analysis of the ascites(HepA) cells, it...
