Objective To develop a novel vector system, which combines the advantages of the gene therapy and virus therapy, and to observe its therapeutic effect on lung cancer. Methods A novel gene-viral vector system, designated CNHK500-hTRAIL, was constructed, in which the E1A and E1B gene was promoted by hTERT and HRE, respectively. We observed its anti-tumor effect and safety on lung cancer therapy by a series of experiments in vitro and in vivo. Results The human TRAIL gene was cloned, CNHK500-hTRAIL was constructed, recombined, amplified, and purified successfully. CNHK500-hTRAIL can replicate and proliferate in lung cancer cells but not in normal cells, thus specifically kill the cancer cells. It introduces the expression of TRAIL gene in tumor cells more effectively than that of traditional gene therapy, inhibits the growth and metastasis of cancer cells, induces cancer cell apoptosis, but doesn't show cytotoxicity on normal cells and mouse liver function. Conclusion Oncolytic adenoviral vector carrying human TRAIL gene holds great promising in cancer therapy.
