| BACKGROUND : Lung cancer is one of the leading causes of death in theworld and the underlying cause for lung cancer has been attributed to various factors including alteration and mutation in tumor suppressor genes. Employing suppression subtractive hybridization (SSH), a gene that is highly expressed in chemosensitive tumor cells was isolated and designated as ClOl. As a putive tumor suppressor gene, ClOl locates on 6q23-25 which is a major lung cancer susceptibility locus. The aim of this study is to test if ClOl gene is able to inhibit tumor growth of lung cancer xenograft.METHODS: The recombinant pcDNA3.1-C101 plasmid was constructed andidentified by restriction endonuclease mapping and sequencing. The targeted cancer cell line that expressed ClOl at lower level was screened and then was inoculated s.c. at the right scapular of nude mice. When the xenograft grew up to 0.5 cm in diameter, the ClOl gene therapy (pcDNA3.1-C101:liposome complex) was processed and observation was followed. HE staining, immunohistochemistry, TUNEL staining and Western blot were applied to elucidate inhibitory effect of ClOl gene on lung tumor xenograft. |
