With the increasing development of modern analytical techniques, fingerprint of Chinese Traditional Medicine (TCM) have come into being. The so-called fingerprint refers to certain TCM are treated properly, and application of some analytical methods to obtain characteristic, which can represent the TCM. Today fingerprint have becoming more and more hot in research of TCM quality and been widely recognized home and abroad. But we must soberly realize that nowadays there still exist obvious deficiency and shortcoming in fingerprint , namely, fingerprint and pharmacodynatics are seriously out of joint. All those are against the original intention and restrict the development and application of fingerprint. Information resulted from fingerprint should be corresponded or highly correlated with pharmacodynatics. This is the issue that would be considered and resolved in thestudy. In this paper, shatianyou (Citrus grandis (L.) Osbeck var. shatinyu Hort.) was studied and the fingerprint was established. Moreover, antitussive and expectorant experiments were conducted to obtain the quantitative characteristic and the data of pharmacodynamics. On the basis of that, grey relational analysis was applied to findthe correlation between shatianyou fingerprint and pharmacodynamics. And we have already achieved such achievement as follows:l.The establishment of shatinyu fingerprint and studies on pattern classificationHPLC was introduced to analyze shatianyou samples from different habitats. Mobile phase, chromatographic column and detected wavelength etc. were optimized to complete the systemic methodology and establish the HPLC fingerprint of shatianyou. What's more, the similarity evaluation of chromatograms was processed by computer aided interpretation. The quantitative fingerprint characteristic was treated with cluster analysis. Then Canonical discriminant function and Fisher's discriminant function were founded, which proved to be consistent with facts. As a result, the method should be of great reliability. Actually, the determined samples just need to be analyzed with the chromatographic conditions described above, and input the quantitative fingerprint characteristic of the samples into computer to calculate, then the classified results would be quickly acquired, which has both academic significance and applied value.2. Studies on the chemical constituents of shatianyouGC-MS was used to analyze the volatile oil of shatianyou including phenol,2-methyl-5-(l-methylethyl),D-Limonene,P-Caryophyllen,a-Bergamotene,p-M yrcene,a-Caryophyllen, Nootkatone etc., which in some degree provides basis for the development and utility of shatinyu.Three 7-methyloxy-coumarin such as meranzin hydrate, pranferin and isomeranzin were isolated from shatianyou and purified by silica gel column. In addition, meranzin hydrate and isomeranzin were firstly isolated from shatianyou, and pranferin was the derivation of meranzin hydrate.3.Study on the correlation between characteristic of Shatianyou fingerprint and pharmacodynamicsAt the premise of the acquired characteristic of fingerprint and pharmacodynamic data, grey relational analysis was selected to show what the constituents represented by fingerprint contributed to pharmacodynamics. It can be concluded that antitussive and expectorant effects are the total results of chemical constituents group in shatianyou. According to the relational grade, the sequence of contribution to antitussive effect is, narigin > meranzin hydrate > bergapten > bergamotene > isomeranzin > naringenin > neoheperidin > prunasin ; while to expectorant effect is, prunasin > neoheperidin > narigin > bergapten > meranzin hydrate>naringenin> isomeranzin> bergamotene. The research has set up a newly comprehensive control pattern to integrate and correspond characteristic of fingerprint and pharmacodynamics, which filled the blank in our country in the field of Chinese Traditional Medicine quantitative relation between fingerprint and pharmacodynamics. In a word, the study has been qualified theoretical meaning and practical value, simultaneously leading the way for application of fingerprint in Traditional Chinese Medicine industry.
