Objective: to investigate the possible role of interstitial cell of Cajal and abnormal signal transduction in smooth muscle cell in the pathogenesis of diabetic gastroparesis and the protective effect of "xiaopifang" .Methods: ①healthy male SD rats were randomly divided into 5 groups:normal ,model ,low dosage,middle dosage and high dosage group,diabetes was induced by IV of alloxan,50mg/Kg except for normal group,and the latter three groups were treated with Chinese herb xiaopifang intragastrically once a day from the beginning to the end of the trial; ②gastric emptying was measured by method of nutritious semisolid paste;③the quantity of cAMP in gastric smooth muscle cell was measured with radioimmunoassay; ④the ultra structure of interstitial cell of Cajal was observed through electron microscope;⑤c-kit positive cells were quantitively measured withimmunhistochemistry assay and computer image analysis system; ⑥c-kit, Gs a and Gi a mRNA positive cells were quantitively measured with in situ hybridization and computer image analysis system respectively.Results:?gastric emptying rate of the normal ,middle and high dosage groups were significantly higher than that of model group(p<0.05 ) respectively,and there is no markedly difference among normal,middle and high dosage groups (p>0.05) ;@the quantity of cAMP of the normal ,middle and high dosage groups was significantly lower than that of model group (p<0.05) respectively;(3)changes of the ultrastructure of interstitial cell of Cajal: the number of the gap junctions between Cajal and nerve cell ,smooth muscle cell were decreased significantly,and the structure of the gap junction were also damaged ,mitochondrion was swelling, vacuoles, the number of organelle was decreased, which all were seen in the model group,but the change of Cajal in the middle and high groups is slightly,which was similar to the normal group;(4)the number of c-kit, c-kit mRNA positive cells of the normal ,middle and high dosage groups was significantly higher than that of model group (p<0.05) respectively;?the expression of Gs a mRNA in the model group was significantly higher than that of the normal ,middle and high dosage groups(p<0.05) respectively;?there was no markedly difference for the expression of Gi a mRNA among 5 groups (p>0.05 ) . |
