A Systematic Comparative Study Of Main Chemical Composition Of Northeast Sika Deer (Cervus Nippon Hortulorum) Velvet

Main chemical composition of Northeast sika deer (Cervus nippon hortulorum) velvet antler was analyzed in this study. In Experiment I, amino acid and other nutrient contents of different slices prepared from different parts ie. tip, upper, upper mid and base of velvet antler, namely wax slice (WS), powder slice (PS), blood slice (BLS) and bone slice (BNS) were determined. Calcium contents of WS, PS, BLS and BNS were 0.31, 4.63, 5.32 and 6.56%, respectively; phosphorus contents were 0.77, 4.89, 5.44 and 6.61%, respectively; magnesium contents were 0.54, 1.37, 1.66 and 1.89mg/g, respectively; potassium contents were 6.62, 4.48, 4.00 and 2.17 mg/g, respectively; sodium contents were 8.46, 7.75, 7.24 and 6.73 mg/g, respectively. The contents of each above element in different parts of velvet antler were, mostly, differed significantly with a few being similar. The contents of calcium, phosphorus and magnesium increased gradually from the tip to the base, which were in accordance with the conclusion reported in literature. The contents of potassium and sodium were decreased gradually from the tip to the base, which were conflict to the conclusion reported in literature. For micro-elements, copper contents of WS, PS, BLS and BNS were 0.24-4.31, 0.65-5.94,0.45-7.65 and 0.50-3.05 Mg/g, respectively; zinc contents were 0.06-0.10, 0.06-0.13, 0.07-0.20 and 0.06-0.10 mg/g, respectively; iron contents were 0.23-0.77, 0.16-1.39, 0.12-0.94 and 0.14-0.34 mg/g, respectively; and manganese contents were 2.89-13.68, 5.46-18.88, 3.16-11.02 and 3.54-12.61 Mg/g, respectively. No trends were found in the contents of the four elements in different parts of antler. ,Water soluble protein contents of WS, PS, BLS and BNS were 3.97,3.05,2.55 and 1.59%, respectively; hydrophobic amino acid contents were 64.14, 54.69, 49.24 and 43.68%, respectively;free amino avid contents were 1.96, 1.47, 1.24 and 0.62%, respectively. Total saccharide contents were 3.21, 2.21, 1.04 and 0.49%, respectively; reducing saccharide contents were 0.24, 0.14, 0.11 and 0.05, respectively; polysaccharide contents were 2.97, 2.07, 0.93 and 0.44%, respectively; phospholipid contents averaged 5.14, 2.48, 1.75 and 1.10%, respectively. Each above component in tip of velvet antler was significantly higher than upper (P<0.01), which, in turn, was significantly higher than upper mid (p<0.01), and which, in turn, was significantly higher than base (p<0.01). These results were in accordance with the results found in literature.Combination of above results can be used as an internal index to evaluate the quality of different parts of sika deer velvet antler. Two activate index programs of Northeast sika deer velvet antler has been designed.In experiment II, chemical composition of petrol ether, diethyl ether, methanol and 80% methanol extracts of Northeast sika deer and rein deer velvet antler and Northeast sika deer antler base were compared. Petrol ether extract was separated with thin layer chromatography using CMC-Na as fixed phase and benzol and diethyl ether (7:1) as mobile phase. The results showed that the components of the extracts of antlers and base differed significantly; while the components of two kind antlers were similar but there were significant differences in the quantity of each component between the two origins of antlers. This pattern might be used for identifying the origin of antler products. The components of diethyl ether extracts of three samples mentioned above were similar when they were separated using the same conditions used for petrol ether extract separation. There were only quantity differences between samples. There were significant differences in the components of methanol extracts of the 3 samples separated with thin layer chromatography usingCMC-Na as fixed phase and cyclohexane, chloroform, methanol and ice acetic acid (20:5:5:0.1) as mobile phase.
