Objective: To explore the mechanisms of neocortex and hippocampus senescence in SAMP 10 and the improvement effects of learning and memory disturbance by "YiQiTiaoXue, FuBenPeiYuan" acupuncture method.Methods: Using Senescence Accelerated Mouse (SAM) P10 and normal control SAMR1 as model, applying the cDNA Array technique, and monitoring the expression difference of 588 genes in seven groups, viz.S-month SAMR1 neocortex and hippocampus control groups, 8-month SAMP 10 neocortex and hippocampus control groups, 8-month SAMP 10 neocortex and hippocampus "YiQiTiaoXue, FuBenPeiYuan" acupuncture method groups, 8-month SAMP 10 hippocampus non-points group.Results: (1) 17genes' expression was different between the 8-month SAMR1 neocortex control group (SAMRl/N group) and 8-month SAMP 10 neocortex control group (SAMP10/N group). 15 genes' expression was down-regulated and 2 genes' expression up-regulated in SAMP10/N group compared to SAMRl/N group. 10 genes' expression was different between the 8-month SAMR1 hippocampus control group (SAMR1/H group) and 8-month SAMP10 hippocampus control group (SAMP10/H group). Sgenes' expression was down-regulated and 2 genes' expression up-regulated in SAMP10/N group compared to SAMRl/N group. Genes with changed expression included: transcription, intracellular transducers/ effectors / modulators, stress response proteins, DNA synthesis/recombination/and repair, cytoskeleton/motility proteins, apoptosis associated proteins, cell cycle, cell signaling, extracellular communication proteins, cell receptors and so on. (2) 20 genes' expression was different between 8-month SAMP 10 noecorex "YiQiTiaoXue, FuBenPeiYuan" acupuncture method group (QiXue/N group) and SAMP10/N group. 15 genes' expression was up-regulated and 5 genes' expression down-regulated in QiXue/N group compared to SAMP10/N group. 16 genes' expression was different between 8-month SAMP10 hippocampus "YiQiTiaoXue, FuBenPeiYuan" acupuncture method group (QiXue/H group) and SAMP10/H group 10 genes1expression was up-regulated and 6 genes' expression down-regulated in QiXue/H group compared to SAMP10/H group. Genes with changed expression included: transcription, intracellular transducers/effectors/modulators, stress response proteins, DNA synthesis/recombination/and repair, cytoskeleton/motility proteins, apoptosis associated proteins, cell cycle, cell signaling, extracellular communication proteins, cell receptors, DNA binding and chromatin proteins and so on. The genes whose expression was different between SAMP 10 group and SAMR1 group was beneficially regulated by "YiQiTiaoXw, FuBenPeiYuan" acupuncture method, viz up regulated the lower genes expression, down regulated the higher genes expression (3) 6 genes' expression was different between 8-month SAMP 10 hippocampus non-points group (Non-points/H group) and SAMP10/H group. 5 genes' expression has the same regulation with QiXue/H group Genes with changed expression included: transcription, cell cycle and cell signaling/extracellular communication proteinsConclusion: The results show that "YiQiTiaoXue, FuBenPeiYuan" acupuncture method can ameliorate the syndrome of learning and memory disturbance in SAMP 10 as a whole.
