Objective: To measure and compare the TF level and the levels of other biomarkers to evaluate the clinical potential of tissue factor as a marker for the diagnosis of deep vein thrombosis after total knee arthroplasty.Methods: Eighty-eight patients who underwent total knee arthroplasty were recruited from Jonit surgery of the Affiliated Hospital of Qingdao University Medical College between May 2014 and March 2015. Patients were randomly divided into two groups according to thrombosis formation: non thrombosis group and thrombosis group. Forty-seven people who underwent normal body check were included in control group. The mRNA level of TF was determined by Real-time PCR. The plasma levels of TF, TFPI, TAT, IL-6, IL-8 and P-selection were measured by ELISA method.The plasma level of D-dimer was determined by automatic biochemical analyzer. Finally compare the change of index of deffrent groups.Results: 1. There was no significant difference in age and gender between control group, non thrombosis group and thrombosis group(P > 0.05). 2. There was no significant difference in TF mRNA and protein levels between control group and non thrombosis group(P > 0.05). TF mRNA and protein levels were significantly increased in thrombosis group(P < 0.05). 3. TF mRNA and protein level in thrombosis patients had no relationship with their age and gender. 4. There was no significant difference in the levels of TFPI, D-dimer, TAT, IL-6,IL-8 and P-selection between control group, non thrombosis group and thrombosis group(P > 0.05). The levels of TFPI, D-dimer, IL-6, and P-selection were significantly higher in thrombosis group than those in control group or non thrombosis group. 5. Relationship analysis showed that TF level was positively related to TFPI(r=0.713, P=0.000), D-dimer(r=0.531, P=0.000) or P-selection level(r=0.699, P=0.000) 6. TF level was significantly reduced in over-anticoagulation group, as compared to non over-anticoagulation group(P< 0.05).Conclusion: 1. TF expression is upregulated in thrombosis patients, indicating that it may be used as a biomarker for the diagnosis of DVT after total knee arthroplasty.2. TF expression in thrombosis patients had no relationship with their age and gender. 3.TF expression is positively related to TFPI, D-dimer or P-selectin expression in thrombosis patients. 4. TF expression is related to over-anticoagulation, indicating that it may be used as a predictor for the diagnosis of over-anticoagulation.
