Buffalograss is a warm seasonal turf grass, which has been planted in China for about 60 years, since it was introduced into China in the fifty decades in last century. It was assessed to be good lawn grass for its outstanding features such as drought resistance, cold resistance and disease resistance. Especially in the north-east, north-west and North China, buffalograss consumes less water and fertilizer, grows slowly and needs less mowing. But buffalograss seeds have a strong dormancy, which restricts it from high turf quality and wide spread. The study on breaking dormancy of buffalograss seeds is meaningful for the production of buffalograss lawn.This study was carried out on Buchloe dactyloide cv. Zhongping No.1, mainly on the reason of dormancy and how to break it. On the basis of this, identify the proteome which influenced the seed dormancy by two-dimensional electrophoresis and mass spectrometric analysis. The results were as follows:1. The seeds of buffalograss were small. The average length was 1.75 mm, and the average width was 1.01mm, thousand seed weight was 2.0g. The glumes grew in an aggregated way with 2~5 seeds in each glumes. There were pore canals between peristachyum. These pores may be the door which plant absorpts water and air by that.2. The result of observation shows that the surface of glumes is bright, compact and mounted with scaly Si-rich matter,the percentage of mass is 26.92% for Si .There were no pores on the scare when the mosaic dropped off, which was different from zoysia glumes. by electron microscope and energy spectrum3. It were certified tha the dormancy type of buffalograss seeds belongde to synthetic type had been dorminated by endogeny facter, with the significant different of germination rate for seeds without glums and seedballs of buffalograss in 2006; the higher viability ingermination seeds in seedballs(more than 80%); the growth diraction of embryonic root; the surface texture and component analysis to glums; the effect that buffalograss seeds had been intivited by GA3 and so on.4. As to break dormancy, dormancy reason, storage life and germination rate after break dormancy should be considered. As to seeds with a storage life more than two years, mechanical method can increase the germination rate to more than 80%. As to seeds with a storage life less than two years or even less than one year, both mechanical and physiological method should be taken for a high germination rate as 80%, for the purpose this, a concentration of 60mg/Kg GA3 was suitable.5.The trizol base method is the best for sample preparation,combination with pH4-7 IPG strip and appropriate time in isoelectric focus (IEF gave rise to satisfactory 2-DE images. The maximal detectable protein spots number were estimated to be more than 800 per gel under silver staining.6. Take the buffalograss seed harvest at 2008 as control, the seed harvest at 2006 and seed harvest at 2008 participation of gibberellic acids(GA) as treated .A total of 34 spots, showing significant changes in volume at least 2-fold, 20 spots of them were identified by MALDI-TOF-MS and 16 spots were successfully identified. The classification of proteins showed that all the proteins associated with dormancy breaking are involved in Energy/electron transport, Photosynthesis, Transcription, Signal transduction and Protein synthesis/ Development.
