This research aimed to investigate the effects of several nitrogenous heterocyclic compounds on soil bacterial diversity in the cornfield and its regulation on the genome of Pseudomonas aeruginosal.The paper discussed the influence of nitrogenous heterocyclic compounds on environmental micro-ecology as well as the decomposition of nitrogenous heterocyclic compounds by related microbes,which helped to illustrate the reaction between nitrogenous heterocyclic compounds and microbes in the molecular level. Moreover it established a solid foundation for the future research of environmental evaluation of the new nitrogenous heterocyclic compounds in the circumstance of lab scale.On the basis of former research,we optimized the synthetic processes of two new nitrogenous heterocyclic compounds-4-amino-6-tert-butyl-3-methylthio-1,2,4-triazin-5 (4H)-one sulfate,4-[2-(4,6-D Methoxy pyrimidine]-3-thiourea Carboxylic Acid phenoxy Ester,and acquired a high yield of product.The bioactivity research indicated that the nitrogenous compounds possessed stable bioactivity which makes it possible to develop new pesticides with intellectual property rights belongs to China.The traditional plate-cultivation method and fluorescent labeled restriction fragment lengthy polymorphism were introduced to investigate the influence of three tested compounds:4-amino-6-tert-butyl-3-methylthio-1,2,4-triazin-5(4H)-one sulfate, N-(4,6-D Methyl pyrimidine-2-) aniline,4-[2-(4,6-D Methoxy pyrimidine]-3-thiourea Carboxylic Acid phenoxy Ester soil on bacterial diversity in cornfield.The results showed less negative effects on the soil bacteria,and good compatibility.It also has the ability to induce the resistant bacteria to the tested compounds Therefore, it's easier to obtain enriched,accurate and reliable data through combination of traditional plate-cultivation method with fluorescent labeled restriction fragment lengthy polymorphism technique.It's also a valid method to evaluate the impact of pesticides on the micro-ecology.It was the first report that Pseudomonas aeruginosa random promoter constructed by the vector pMS402 could be used to screen the tested compounds Seven promoter related to degradation of the tested compounds were obtained by screening the libraries of three nitrogenous compounds-4-amino-6-tert-butyl-3-methylthio-1, 2,4-triazin-5(4H)-one sulfate N-(4,6-D Methyl pyrimidine-2-) aniline, N-(3-ethyl-5-methyl-4H-1,2,4-triazol-4-yl)(4-fluorophenyl) methanimine.The genetic maps related to individual functional genes were constructed after sequencing and aligning.The initial researches proved that two functional genes definitely encoded dehydrogenate kinase of acetyl coenzymes and phosphate transferase respectively,others are presumably pertinent to componential proteins of transport system,membrane proteins,redox proteins and dual-component regulators.The technology implies a new approach to the microbial decomposition of tested compounds...
