The central section in petiole of Osmanthuare was anatomized, and it was discoveredthat the number of faseicular was mostly 1 large and 2 small, the type of fascicular hadamphicribral bundle, almost amphicribral bundle and collateral bundle, and the marrow cavitywas only found in the center of vascular bundle of exceptional species, In one word, structurecharacteristics of central section in petiole of Osmanthus were rather stable and wereconsidered to be the representative characteristics, and the evolvement trend of vascular bundlein vein was amphicribral bundle, almost amphicribral bundle and collateral bundle.6. The MP, NJ and UPGMA phylogeny trees constructed according to 22 kinds ofcharacteristics of Osmanthu were basically similar, and which was identical with viewpoint oftradition taxology. Although MP, NJ and UPGMA phylogeny trees resulting from 45 kinds ofcharacteristics of Osmanthuare were diverse with viewpoint of tradition taxology,distinguishment of relative between some species held out viewpoint of tradition taxology.7. It was firstly found that the length of ITS, ITS-1, ITS-2 and 5.8S of Osmanthus wasseparately 245bp, 220bp, 628bp and 163bp. The G+C content of ITS-1, ITS-2 and ITS was54.24-64.02%, 54.63-63.43%, 53.98-61.20%, respectively. In addition, the aberrant points andinformative points of rDNA ITS sequence in Osmanthus plant were 198 or 97, separately, inwhich there were 94 aberrant points and 47 informative points in ITS-1, and 92 aberrant pointsand 47 informative points in ITS-2. Moreover, there were 12 aberrant points and 3 informativepoints in 5.8S. So, it was illuminated that the heredity diversity of Osmanthus could be studiedby ITS sequence because ITS sequence of Osmanthus generally exhibited polymorphic,whereas the clustering results by ITS sequence was different from viewpoint of traditiontaxology, some of which could sustain viewpoint of tradition taxology.8. Although ISSR analysis results were diverse with viewpoint of tradition taxology, partly sustained viewpoint about interspecific relations from tradition taxology. In addition, there werelargely consistent between clustering results of ISSR and ITS of Osmanthus, showing that itwas feasible and effective to research the classification of Osmanthus by DNA molecularmarker.
