Using young panicle of a haploid rice as explant, a tissue culture assay was carried out. Regeneration plants were obtained by callus induction and redifferentiation. The results indicated that: the haploid young panicle could diploidize in the course of tissue culture; the fertility of double-haploid pollens was good; the caryopsises of double-haploid was red. The SSR technique showed that the progenies of parts of double-haploid after self-crossing were homolog. Foremostly, the red caryopsis rice was discovered.Reciprocal crosses between red and achromatic pericarp rice revealed that the seed color of F1 was determined by its female parent. According to the caryopsis pericarp color and plant segregation ratio of F1, F2, and F3 generations, the red phenotype of red double-haploid (rdh) caryopsis was determined by a dominant, monogene with maternal effect. Histological study showed that the red pigments accumulated in the pericarp layer only. The assay of developmental timing of pigment accumulation showed that the red color accumulated from desiccation stage to perfectly maturation stage of the caryopsis.rdh plant had purple stigmas. Genetic analysis suggested that the purple stigma trait of rdh was controlled by a pair of dominant nuclear genes. Polymorphic analysis of microsatellite markers demonstrated that the purple stigma gene of rdh was located on rice chromosome 6, 4.2cM, 0.35cM and 0.53cM from microsatellite markers RM276, RM253 and RM111, respectively. It was believed that purple stigma gene of rdh was the first mapped purple stigma gene on rice chromosome 6. This purple stigma gene was designated tentatively as Ps-4.Polymorphic analysis of microsatellite markers demonstrated that the red pericarp gene of rdh was located on rice chromosome 7, 4.5cM and 10.3cM from microsatellite markers RM214 and RM320, respectively. It was believed that red pericarp gene of rdh was the first mapped red pericarp gene on rice chromosome 7.
