Through molecular biological technique in combination with morphological characteristics, we, for the first time, made molecular ecological research of seven populations of Sympegma regelii, Kalidium foliatum, and Salsola passerina belonging to Chenopodiaceae in Gansu, Qinghai, Ningxia and Inner Mongolia by using ISSR marker and nrDNA ITS sequence, in order to contrast the genetic mutation within and between populations, to discover the effects that environmental elements have on the genetic diversity, to understand the geographical structure of these species systematically, to probe into the relationship between genetic diversity and molecular phylogeographic structure of these species, to investigate their expanding routes. The final aim is to disclose the influence of abiotic factor on their genetic diversity within and between population in order to get the genetic proof at molecular level for the conservation of the genetic resources of these desert species and improvement of ecological environment in the cold and arid wilderness.In this study, plant material of 417 individuals from seven populations in Gansu, Ningxia, Qinghai and Inner Mongolia were collected for analysis by using molecular marker technology of ISSR (Inter simple sequence repeats) and nrDNA ITS sequence. In addition, 42 soil samples in the habitat sites of these species were obtained by measuring soil moisture, salinity, PH value and major nutrients as well as organic matter.For the 139 individuals of Sympegma regelii from the seven populations, 100 ISSR primers were screened, among them 16 primers produced a total of 212 replicated bands, of which 169 were polymorphic.
