The parthenogentic activation and development in vitro of mouse ooctyes, the mature culture of oocytes in vitro,parthenogentic activation and development of goat oocytes were analysed.At the base of above,the interspecies nuclear transplantation experiment between mouse and goat were made (the mouse/goat granulosa cells used as donor cells,the enucleated MⅡ oocytes of mouse/goat used as cytoplasma recipient). The results and conclution can be found as follow:1 The parthenogentic activation of mouse oocytes1.1 Matured oocytes of mouse activated by 7% ethanol(6min,7min and 8min),then cultured them in three medium(CZB,KSOM and M199).The results showed that: the activation rate was increase along with the delay of activation time.Meanwhile, the cataclasm rate of oocytes was increase. The activative rate of 7min and 8min group was higher than 6min group(P<0.05), no significant different between 7min and 8min group. It was said that the suitable activation time was 7min.1.2 The activation of mouse oocytes was studied by use of ionomycin united with 6-dimethyladenine(6-DMAP).Then, the actavited oocytes were cultured in three medium(CZB,KSOM and M199). The results showed that: the activation rate of mouse oocytes had increased signicantly, the cataclasm rate increased along with the delay of activation time too.It was said that the suitable activation time was 4h..1.3 The 7% ethanol(7min) activated oocytes of mouse were cultured in three medium(CZB,KSOM and M199).The results was made as follows:The highest cleavage rate was CZB(71.2%),secondly was KSOM(65.4%),the lowest was M199(53.3%).There had a significant different between CZB and M199(P<00.05). The highest blastocyst rate was CZB(23.8%),the lowest was M199(7.0%), and the KSOM(17.1%) was the middle among them.It was said that CZB was the best mudium of activated ooctyes of mouse.1.4 The ionomycin(5min) united with 6-dimethyladenine(6-DMAP)(4h) activated oocytes of mouse were cultured in three medium(CZB,KSOM and M199).The results was made as follows:The highest cleavage rate was KSOM(82%),secondly was CZB(80%),the lowest was M199(75.4%).There were no significant different among them(P>0.05). The highest blastocyst rate was CZB(32.5%),the lowest was M199(12.3%), and the KSOM(27.2%) was the middle among them.It was said that CZB and KSOM were all fit for the cluture of the activated oocytes of mouse.2 Mature culture of goat oocytes in vitro2.1 The matured rate(66.2%,72.2%,68.8% and 71.5%, respectively) of goat oocytes in vitro was increased significantly by added 5%/10% fetal bovine serum (FBS) or 5%/10% estrous goat serum (EGS) in the base medium of M199. It was said that the serum was a important influence factor to the mature of oocytes of goat in vitro. 2.2 In the base medium of M199+10%FBS, after added different hormone and foctor(FSH,LH,EGF,HMG), the mature rate of goat oocytes was increased slightly in vitro.It was said that the add of hormone and growth facotr was fit to mature of goat oocytes.3 The parthenogentic activation of mouse oocytes3.1 Goat oocytes was activated with room temperature(25℃) for 3h,4h and 5h,the cleavage rate was 2.5%,4.2% and 2.9% respectively, and the blastocyst rate was 0%.It was said that the temperature activation method was not fit for goat oocyte.3.2 Although 7% ethanol united with 6-dimethyladenine(6-DMAP) could partly activate oocytes of goat, but the blastocyst rate(0%-5.2%) was lower.So,this method was not fit for activation of goat oocyte.3.3 The ionomycin(5min) united with 6-dimethyladenine(6-DMAP) activation(3-5h) could abtain higher cleavage rate and blastocyst rate.The highest blastocyst rate was 43.0% by used of ionomycin(5min)+ 6-DMAP-CB(4h) method.Interspecies nuclear transplantation and early development of embryo between mouse and goat4.1 In the ionomycin(5min)+ 6-DMAP-CB(3h,4h,5h) method,the cleavage rate and blastocyst rate of nuclear transplantation embryos were influenced greatly by activation time.
