The effects of parthenogenetic activation and cytoplasm elimination on development of goat parthenogenetic embryos were analyzed before nuclear transfer. Then, human embryonic fibroblasts cultured in vitro were transferred into enucleated goat oocytes, in order to investigate whether human somatic cell can be reprogrammed in the oocytes of goat and whether the human-goat interspecies reconstructed embryos can develop in vitro. Methods1. Isolation and culture of human embryonic fibroblasts in vitroHuman embryonic fibroblasts were obtained from 5~9 weeks human embryos through enzyme digestion and selected by cell attachment. Morphology of human embryonic fibroblasts was observed, and cell growth curve was made with MTT. Karyotype of human embryonic fibroblasts in passage 10 was analyzed, and cell cycle was done by flow cytometry after cell stage was synchronized by either serum starvation or contact inhibition.2.
