Purpose:By sorting out the literature on the treatment of nonproliferative diabetes retinopathy with Chinese medicine for more than 20 years,and using the Chinese medicine inheritance assistance platform(V2.5)software,we explored the drug use rules for the treatment of nonproliferative diabetes retinopathy and conducted network pharmacology analysis on high-frequency drug pairs,summarized the academic ideas of the tutor in treating DR,and provided more methods for future clinical treatment.Material and method:1.Literature research:Using CNKI,Wanfang Date,and VIP databases,we searched the clinical observation literature on the treatment of nonproliferative diabetes retinopathy with traditional Chinese medicine from January 1,2001 to December 31,2022,and entered it into Excel to establish a database.Through the TCM inheritance assistance platform,we collected the drug frequency,the nature and taste of meridians,and obtained new prescriptions based on association rules and entropy hierarchical clustering.2.Network Pharmacological Analysis:On the Traditional Chinese Medicine System Pharmacology Analysis Platform(TCMSP),enter high-frequency drug names and screen out effective and active ingredients.And through the relevant disease database to screen out the action targets of NPDR diseases,after intersection with drug targets,protein interaction analysis is performed on the intersection genes to screen out core targets.The "drug component disease target" network diagram is formed using Cytoscape 3.9.1,and then functional enrichment analysis is performed on the intersection targets to predict molecular mechanisms such as biological processes and signal pathways.3.Professor Zuo Tao’s Experience Summary:Through the study of teachers during the master’s period,the academic ideas of Professor Zuo Tao in the treatment of diabetic retinopathy were summarized,effective cases were screened,in-depth analysis was made,and the clinical ideas of Professor Zuo Tao in the treatment of this disease were discussed.Result:1.Literature research1.1 A total of 117 documents meeting the requirements were included,120 prescriptions were used,143 traditional Chinese medicines were used to treat NPDR,and the total frequency of drugs was 1293 times.1.2 The top three traditional Chinese medicine categories are: tonifying yin,clearing heat and cooling blood,and tonifying qi1.3 According to the analysis of the four gases of drugs,the most frequently used drugs are cold and warm.1.4 In the analysis of five flavors,the consumption of sweet drugs is the largest,followed by bitter drugs and sweet drugs.1.5 In the analysis of drug meridians,the liver and kidney meridians are the most frequently used.1.6 High-frequency drug pairs are five groups of drug pairs: astragalus-panax notoginseng,raw rehmannia-astragalus,salvia miltiorrhiza-astragalus,astragalus-angelica,and salvia miltiorrhiza-panax notoginseng.2.Network pharmacology,select astragalus and panax notoginseng to analyze the treatment of NPDR2.1 There are 27 active components of the drug,including 1 common component,and215 drug targets after weight removal.2.2 633 disease targets were obtained.2.3 The core targets are 23,including AKT1,TNF,IL6,TP53,VEGFA,IL1 B,CASP3,MMP9,etc2.4 In biological processes,these genes are mainly involved in the positive regulation of cell migration,the response to inorganic substances,the response to lipopolysaccharide,the response of cells to nitrogen compounds,the response to oxygen levels,etc.In cell composition,it involves vesicular cavity,membrane raft,transcriptional regulatory complex,extracellular matrix,protease inhibitor complex,outer plasma membrane,etc.The molecular function mainly involves cytokine receptor binding,kinase regulatory activity,DNA-binding transcription factor binding,protease binding,protease binding,protein domain-specific binding,protein kinase activity,tumor necrosis factor receptor superfamily binding,growth factor binding,etc.2.5 The KEGG pathway enrichment analysis was carried out on the core targets,and a total of 182 action pathways were identified.The results showed that the pathways closely related to these targets were: the cancer pathway,the role of AGE-RAGE signaling pathway in the complications of diabetes,etc.Conclusion:1.Traditional Chinese medicine is commonly used to treat NPDR with tonifying and clearing away heat drugs,mainly involving the liver and kidney meridians.2.The mechanism of Huangqi Sanqi in treating NPDR involves oxidative stress,inflammation,etc.3.The professor applied the idea of "preventive treatment" to all stages of prevention,treatment-rehabilitation of the treatment of sugar network,regulated the liver,spleen and kidneys,formed a "nourishing the liver and kidneys,strengthening the spleen and invigorating qi",and added acupuncture,direct current iontophoresis,warm Sha and other external treatment methods,which is a system that integrates prevention before disease,specimen coherence,pre-existing disease prevention and degeneration,and post-malaria prevention and treatment of thirst and eye disease.NPDR is the initial stage of DR,qi and yin deficiency as the basis,dry heat as the standard,the tutor in clinical application on the basis of nourishing liver and kidney on the basis of nourishing yin and clearing heat,consistent with the theory of literature research.
