Exploration Of Medication Rules And Mechanisms In Professor Yu Rui's Treatment Of Insomnia Of Liver Depression And Phlegm Heat Typ

Objective:Based on data mining and network pharmacology,the medication rules and core formulas of Professor Yu Rui in the treatment of hepatic depression and phlegm fever insomnia were summarized,and its mechanism of action was discussed.Data and methods:Professor Yu Rui’s outpatient treatment of hepatic depressed sputum fever insomnia from October 2020 to March 2022 was collected and the data was imported into the "Ancient and Modern Medical Case Cloud Platform" system to basic information analysis,association rule mining,cluster analysis and co-occurrence network visualization.the medication rules and core drugs of Professor Yu Rui in the treatment of hepatic depressed sputum fever insomnia were summarized.A complex network was constructed for the Chinese medicines prescription obtained by data mining in the previous step,and the active ingredients and corresponding targets of the prescription were found in the TCMSP database.TTD,Dis Ge NET,Gene Cards disease database for disease target gene search,looking for disease and synthetic intersection target genes.The STRING database was used to construct a PPI network to find the core target genes and active ingredients of hepatic depressed sputum fever insomnia.Using the DAVID database,enrichment analysis was performed on intersecting target genes to explore the mechanism of action of the synthesis.Results:In the data mining part,a total of 210 medical cases were analyzed,and 207flavors of traditional Chinese medicine were used,with a total frequency of 4828 uses.The frequency of use in the top 10 are:Banxia,Gardenia,Divine Comedy,Chaihu,Xiangfu,Cangshu,Angelica,Poria,Skullcap and Chuanxiong.The top 5 efficacy of traditional Chinese medicine is to clear the mingmu,relieve liver depression,eliminate irritability,regulate menstruation and relieve pain,dispel wind and relieve pain.The medicinal properties are mainly flat,cold and warm;The medicinal taste is mostly sweet,pungent and bitter;There are more liver meridians,lung meridians,and spleen meridians.Association rules:Banxia-Chaihu-Shuangqin has high support.The complex network diagram combined with the Yu Rui’s experience to formulate the basic formulas are Banxia,Xiangfu,Cangshu,Chaihu,Chuanxiong,Gardenia,Shuangqin,and Shenqu.In the network pharmacology part,the key active ingredients of the prescription were quercetin,stigmasterol,kaempferol,etc.The key intersection targets of drugs and diseases are AKT1,IL6,IL1 B,CASP3,VEGFA,etc.GO function enrichment involves GABA herniation,neuronal apoptosis,etc.KEGG is mainly enriched in the TNF,HIF-1,IL-17 and NF-κB signaling pathway.Conclusions:1.Professor Yu Rui believes that liver depression phlegm fever insomnia is located in the liver,which is closely related to the heart,spleen and lungs,and the basic pathogenesis is liver qi stagnation,phlegm fire endogenous,to relieve liver depression,phlegm and heat as the treatment principle,commonly used prescription is Xiao Chai Hu Tang and Yueju Maru.2.Network pharmacology research proves that Professor Yu Rui’s core prescription drug for the treatment of hepatic depressed sputum fever insomnia has a multi-component,multi-target and multi-pathway joint effect.Active ingredients such as quercetin,kaempferol,and stigmasterol act on target genes such as AKT1,IL6,IL1 B,CASP3,MAOA,VEGFA,etc.,and regulate signaling pathways such as AGE-RAGE,IL-17,TNF,HIF-1,and NF-ΚB to treat insomnia.
