Objective:(1)Metabolomics was used to explore the characteristics of metabolites in myocardial tissue in rats with coronary heart disease(CHD)in combination with heart Qi deficiency syndrome and to explore the possible biological mechanisms of traditional Chinese medicine(TCM)syndromes.(2)To detect specific metabolites and related pathways in the myocardium of model rats following treatment with Yangxin Tongluo Formula,to explore its potential mechanism,and provide guidelines for clinical treatment.Methods:Forty rats were randomly divided into four groups:the control group,and the model group of CHD with heart Qi deficiency syndrome,the TCM treatment group,and the.metoprolol tartrate(MET)group.Thirty rats were modeled according to following methods:controlled feeding,forced heavy-load swimming,and high-dose administration of Xindean.Specifically,the rats underwent continuous exhaustive swimming for 21 days,oral gavage of Xindean for 4 days,and subcutaneous injection of isoproterenol for 5 days.After model validation,the rats were divided into three groups:the model group,the TCM group,and the MET group.The rats in TCM group received oral gavage of TCM for 14 days.The rats in MET group were intragastrically given MET suspension for 14 days.At the end of experiment,all rats were killed and the tissues were sampled.UPLC-MS was used for non-targeted metabolomics analysis,and multivariate and univariate statistical methods were used to identify differentially expressed metabolites and screen for potential biomarkers.KEGG,MSEA,topology,and trend analysis were used to identify differential metabolic pathways.Results:(1)Myocardial metabolomics in the CHD heart qi deficiency syndrome model group:A total of 115 different metabolites,8 potential biomarkers,35 metabolic sets,and 12 metabolic pathways were identified between the model group and the normal group.(2)Myocardial metabolomics in the TCM group:A total of 41 differential metabolites,5 potential biomarkers,1 metabolic set,and 1 metabolic pathway were identified between the TCM group and the model group.(3)Myocardial metabolomics in metoprolol tartrate group:A total of 81 differential metabolites,8 potential biomarkers,9 metabolic sets,and 6 metabolic pathways were identified between metoprolol tartrate group and the model group.Conclusions:The myocardial metabolism of rats of CHD with heart Qi deficiency syndrome showed related specific metabolites.The metabolites changed after intervention with the TCM,indicating that TCM exerts its effects by regulating metabolic pathways.Further research requires clinical samples to validate the above results. |
