Objective:By intervening with Penyan Pill in rats with sequelae of pelvic inflammatory diseases,the mechanism of action of Penyan Pill on SPID model rats was explored from the general condition,morphology of uterine tissue,pathological changes,changes in serum iron ions,and Nrf2 and HO-1 mRNA levels in tissues,so as to provide a theoretical basis for its clinical application and provide direction for more in-depth research.Methods:Ten of the 40 SPF-rated female SD rats were randomly selected as the sham group.The rest of the rats were established with SPID rat models and verified the modeling results,and after successful model verification,they were randomly divided into model group,Penyan Pill group,and azithromycin group,with 9 animals in each group.Gastric gavage distilled water in the sham group and model group,corresponding to the gavage liquid in the Penyan Pill group and azithromycin group,continuous gavage for 4 weeks.Record and evaluate the general condition of the rat and the general morphology of the rat uterus;Uterine tissue structure,inflammatory cells,fibrosis and endometrial glands were observed with HE staining.Detection of serum iron with Elisa;The expression of Nrf2 and HO-1mRNA in uterine tissue was detected by RT-qPCR.Results:The general view of uterine morphology of rats in the Penyan Pill group was close to that of rats in the sham group,and there were significant morphological differences compared with rats in the model group.The pathological section of the uterus of rats in the Penyan Pill group showed that the uterine tissue structure was clear,a small amount of inflammatory cells in the lamina propria,and the morphology of the endometrial glands was normal,which was significantly improved compared with the model group.By detecting the serum iron content of rats,iron death occurred in SPID model rats,and the serum iron content of Penyan Pill rats decreased significantly compared with the model group(P<0.05).By detecting the expression of Nrf2 and HO-1 mRNA in rat uterine tissue,the results showed that the Peny an Pill group had higher expression(P<0.05).Conclusion:1.Penyan Pill can alleviate the symptoms of SPID model rats,restore the morphology of rat uterine tissue,and reduce inflammation.2.Penyan Pill play an anti-inflammatory role by regulating the expression level of Nrf2 and HO-1 in tissues to reduce serum iron content to reduce the occurrence of iron death in tissue cells.3.There were significant differences between Penyan Pill group and model group in various detection indicators,indicating that Penyan Pill could inhibit uterine inflammation and improve pelvic state in SPID model rats.However,the indicators of the Penyan Pill group have not yet reached the level of the sham surgery group,indicating that Penyan Pill cannot completely repair the damaged tissue to normal levels in the short term.
