| Objective: By observing the effects of Shui Lian Feng Kui Prescription of Yao Medicine on the expression of cytokines interleukin-6(IL-6),IL-8,matrix metalloproteinase-2(MMP-2)and vascular endothelial growth factor(VEGF)in serum of rats with sequelae of pelvic inflammatory disease,the mechanism of its action on SPID and its therapeutic advantages were explored.Methods: 60 female SPF grade SD rats were selected as subjects,and after 7days of adaptive feeding,they were randomly divided into model group,gynecological qianjin tablet group and high,medium and low dose groups of Shui Lian Feng Kui Prescription of Yao Medicine by random number method.Except the blank group,the SPID model was copied by the method of "mechanical injury + mixed bacterial infection".After successful modeling,the blank group and the model group were given equal volume distilled water by gavage,and the other treatment groups were treated with corresponding drugs.After 21 days of intragastric administration,the uterus and its surrounding pelvic appendages were taken for observation and detection.The histological changes of uterus were observed after HE staining.The expression levels of MMP-2 and VEGF proteins in the uterus were detected by Western blot,and the levels of cytokines IL-6 and IL-8 in the serum were detected by enzyme-linked immunosorbent assay(ELISA).Results:(1)morphological observation: by visual observation,the uterus and bilateralfallopian tube tissues of SPID rats in the model group showed obvious congestion and local swelling,and the uterus adhered to the surrounding tissues of the pelvic cavity or there was pelvic effusion.In the blank group,there were no hyperemia,swelling and pelvic adhesion in uterine and fallopian tube tissues.In the other treatment groups,only a small amount of the uterine and fallopian tube tissues were found.HE staining under the microscope showed that the wall structure of the endometrium in the blank group was clear,there was no hyperemia,edema and necrotic shedding,and no inflammatory cell infiltration was observed.In the model group,the endometrium wall structure was changed,the endometrial hyperemia and edema were observed,and a large number of epithelial cells were proliferated,denatured,and necrotic,with a large number of or lamellar inflammatory cells infiltrating,and the gland cells were atrophic and deformed.It suggested that the rat model of pelvic inflammatory disease caused by endogenous pathogen infection was successful.In the high-dose group,the inner wall structure of the uterus was basically clear,the epithelial cells were arranged neatly,a small amount of epithelial cells were denatured and necrotic,the glands were basically intact,a small amount of inflammatory cells were infiltrated,no obvious adhesion was observed,and the serosal layer structure was basically intact.The endometrial inflammation in other treatment groups was less severe than that in the model group and more severe than that in the high-dose group;(2)Effects on serum IL-6 and IL-8 levels: compared with the blank group,the serum IL-6 and IL-8 levels in the model group were significantly increased(P< 0.05),and the expressions of VEGF and MMP-2 proteins in the rat uterus were significantly increased(P < 0.05).Compared with the model group,the contents of IL-6 and IL-8 in serum of the rats were significantly reduced byShui Lian Feng Kui Prescription(P < 0.05);(3)Effects on the expression of VEFG and MMP-2 proteins in the uterine tissue: compared with the blank group,the expression of VEGF and MMP-2proteins in the rat uterine tissue was significantly increased(P < 0.05).Compared with the model group,Shui Lian Feng Kui Prescription could significantly inhibit the expression of VEGF and MMP-2 proteins in rat uterine tissues(P < 0.05);Conclusion:(1)Shui Lian Feng Kui Prescription of Yao Medicine has obvious therapeutic effect on SPID rats,can significantly improve the endometrial tissue hyperemia,edema,pelvic tissue adhesion and other inflammatory state,significantly reduce the necrosis,exfoliation and inflammatory cell infiltration of epithelial cells;For SPID rats,the expression of VEGF and MMP-2 can be inhibited by down-regulating the serum levels of IL-6 and IL-8,and the symptoms of the sequelae of pelvic inflammatory disease can be improved;(2)the serum levels of pro-inflammatory cytokines such as IL6 and IL-8 were decreased in SPID rats,which was considered to inhibit the synthesis or release of pro-inflammatory cytokines,thereby reducing the inflammatory reactions such as tissue congestion and edema,and promoting the repair of inflammatory tissues;... |
