Study On The Rules, Mechanism And Efficacy Of Traditional Chinese Medicine In The Treatment Of Liver Meridian Stagnation And Heat Type Acne Mastitis Based On Big Data And Clinical Trial

Purpose:Through the application of data mining technology in the era of big data,to study the rule of prescription for treating liver meridian and pyretic acne mastitis in recent 20 years,and to obtain the new core prescription;By using the network pharmacology technology,the mechanism of action of the core prescription in treating liver meridian heat-stasis type comedogenic mastitis was explored,and its therapeutic effect was observed through clinical trials,providing basis for clinical treatment of liver meridian heat-stasis type comedogenic mastitis and future experimental research.Methods:Through series of China know net database(CNKI),Chinese periodical service platform(VIP),the data of ten thousand literatures about knowledge service platform for nearly 20 years,a record medical record data by using Excel software and prescription of mining frequency,category,sexual flavour to the analysis,using the correlation analysis and SPSS software system clustering analysis,Get the core components.TCMSP database and CNKI database were used to investigate the effective components and corresponding action goals of various traditional Chinese medicine recipes.The disease target of plasma cell mastitis comes from the Gencard database,and the common goal of disease and drug is from the Venny website.With Cytokeratin 3,a network of "disease resistance targets of using Metaatlas database,the target route network map was created and the important routes and main destinations were selected.A total of 30 patients with acne mammary carbuncle were randomly divided into observation group and control group,with15 cases in each group.Both groups were treated with mammary perfusion 3 times a week for4 weeks.In addition,the observation group was additionally treated with Chinese medicine decoction supplemented by the core prescription,100 ml orally three times a day,one week as a course of treatment,a total of 4 courses.The effective rate,the proportion of mass area in the total breast area and the safety of the two groups were analyzed.Results:1.139 literatures related to liver meridian heat-stasis type acne breast carbuncle were included,involving 145 Chinese medicines.The results of frequency analysis showed that the top 10 drugs were Bupleurum,radix paeoniae,dandelion,Angelica,licorice,Scutellaria baicalensis,Hedyotis bungeana,honeysuckle,salvia miltiorrhiza and forsythia.The results of classification and taste showed that heat-clearing drugs,blood-activating and stasis-removing drugs were more commonly used.Taste belongs to cold,warm,flat;Bitter,spicy,sweet;And mostly in the liver,spleen,stomach and other viscera.Correlation analysis showed that support the higher medicine of radix bupleuri,radix scutellariae,radix bupleuri-spreading hedyotis herb,radix bupleuri,salvia miltiorrhiza and radix bupleuri-spreading hedyotis herb,radix paeoniae rubra,radix bupleuri,salvia miltiorrhiza,red paeony root,etc.;The drug pairs with high confidence were Bupleurum-Tuber,Bupleurum-gardenia,Scutellaria baicalensis,Bupleurum-polygonum cuspidatum,Bupleurum-salvium-liquorice,Bupleurum-polygonum cuspidatum,bupleurum-gardenia officinalis and paeoniae paeoniae.The first category of prescriptions included hawthorn,Polygonum cuspatum,Hedyotis bungeana,Paeony root,salvia miltiorrhiza,dandelion,honeysuckle,Trichosanthes,Angelica;The two kinds of prescriptions included: Scutellaria baicalensis,Gardenia jasminoides,Bupleurum chinense,turmeric.The three kinds of prescriptions include liquorice,tangerine peel,forsythia,Prunella sinensis,and saponella spp.The final core components are: Bupleurum,Scutellaria baicalensis,hedyotis bungeana,salvia miltiorrhiza,red peony root,dandelion,gardenia jasminoides,Hawthorn and turmeric.2.Network pharmacology results showed that the main active components of the core prescription in the treatment of plasma cell mastitis were quercetin,luteolin,kaonephol,baicalin,stigmasterol,β sitosterol,etc.The key targets included IL1 B,ESR1,PGR,TNF,IL6,IL4,CXCL8,etc.The main biological processes include negative regulation and translation of megakaryocyte differentiation,cell response to pressure regulation,cell response to chemical stress,cell response to lipid,etc.It is mainly related to the cell composition of riboprotein complex,chromosomal telomere region,focus adhesion,ribosome and vesicle cavity,and mainly acts on the molecular functions of protein domain-specific binding,cell adhesion molecule binding,ubiquitin protein ligase binding,m RNA binding,transcription factor binding and so on.The main enrichment pathways were alcoholism,NF-κB signaling pathway,MAPK signaling pathway,tumor necrosis factor signaling pathway,IL-17 signaling pathway,Toll-like receptor signaling pathway,PI3K-Akt signaling pathway,etc.3.The clinical observation results showed that there was no significant difference between the two groups in age,time of onset and the proportion of mass area in the total breast area before treatment(P > 0.05);After 4 weeks of treatment,the effective rate of the observation group was 93.33% and that of the control group was 60%,P < 0.05.The analysis of the proportion of the mass area in the total breast area after treatment showed that the observation group was(7.37 ± 5.29)% and the control group was(13.77 ± 10.66)%,P < 0.05,which was statistically significant.Conclusion:1.In recent years,the doctor treat liver meridian wind-cold type acne medication prescription has a acne breast carbuncle.The combination of the two is a safe and feasible treatment.
