Objective:1.Using data mining to analyze the medication law of treating idiopathic membranous nephropathy with deficiency of spleen and kidney yang2.Explore the pharmacological mechanism of idiopathic membranous nephropathy by using network pharmacology.Materials and methods:1.Collect Yuan fang professors From September 2019 to November 2021,outpatients who met the CRITERIA of IMN spleen and kidney yang deficiency in the Affiliated Hospital of Liaoning University of Traditional Chinese Medicine used EXCEL software to establish a literature information database,and the data mining methods of drug attribute analysis,association rule analysis,and cluster analysis were used to explore the medication rules of Yuan fang professors in diagnosing and treating IMN spleen and kidney yang deficiency.2.Based on the method of network pharmacology,this paper analyzes the mechanism of the core prescription of Yuan fang professor for the treatment of IMN spleen kidney yang deficiency syndrome,obtains the effective components and matched corresponding targets of the core drugs through tcmsp database and Uni Prot database,and obtains the disease targets in GENECARD database.After using the Wayne diagram to obtain the common target of drug diseases and matching the corresponding gene protein name,enter the string 11.0 database to obtain the target contact network diagram,and use Cytoscape 3 8.2 draw visual diagram.The combination of drug enrichment mechanism of gkeg pathway and target function of gkeg pathway was introduced into the database.Research results:1.The data mining results show that the sex age distribution law is the largest number of IMN patients aged 50-69 years,and the number of female patients is more than that of male patients.The 183 formulas screened out were analyzed,involving 207 flavors of traditional Chinese medicine,and most of the drugs were used to supplement the deficiency of ganping,into the spleen meridian and the kidney meridian.The analysis of 183 formulas involving 207 flavors of traditional Chinese medicine,the analysis of drug frequency shows that there are 27 flavors of commonly used drugs in Wushi,and according to the association rules,3 groups of commonly used drug combinations were excavated by Professor Yuan,and cluster analysis showed 7 drug combinations.Among them,the confidence and support of astragalus-leukotherapy were higher than those of other drugs,so the pharmacological mechanism of astragalus-leukotherapy was analyzed.2.Network pharmacology results: The pharmacological mechanism analysis of the core drug combination astragalus-leukotherapy was carried out,and the active ingredients that played the main therapeutic effect were quercetin(Quercetin)and kaempferol(Kaempferol);92 drug disease interaction targets were screened,mainly interleukin 6(IL6),tumor necrosis factor(TNF),vascular endothelial growth factor(VEGFA),and interleukin 1β(IL1β).It involves podocyte repair and the regulation of inflammatory responses.GO analysis is mainly enriched in biological processes: including positive regulation that affects cell migration,response to inorganic substances,response to hormones,membrane raft,transcriptional regulatory complex,pernuclear region of the cytoplasm,lateral plasma membrane,kinase binding,cytokine receptor binding,etc.KEGG pathway analysis shows that this drug combination may function through the IL-17 signaling pathway and the MAPK signaling pathway.Conclusion:1.Professor Yuanyuan treats the syndrome of deficiency of spleen and kidney yang in idiopathic membranous nephropathy,emphasizing strengthening the right and eliminating evil.Righting is mainly to support Spleen Qi,spleen yang,kidney qi and kidney yang,while dispelling evil is mainly water evil,blood stasis and damp heat,Special emphasis is placed on the role of the spleen.2.The pharmacological mechanism of the core drug combination Astragalus macrocephala Atractylodes macrocephala was analyzed,and two active components were obtained:Quercetin and kaempferol;92 drug disease interaction targets were screened,mainly IL6,TNF,VEGFA and IL1 β。 It involves podocyte repair,and regulation of inflammatory response.Go analysis is mainly concentrated in biological processes: including positive regulation affecting cell migration,response to inorganic substances,response to hormones,membrane raft,transcriptional regulatory complex,perinuclear region of cytoplasm,outer side of plasma membrane,kinase binding,cytokine receptor binding,etc.KEGG pathway analysis showed that this drug combination may play a role through MAPK signaling pathway and IL-17 signaling pathway. |
