Study On The Intervention Effect Of Bushen Huoxue Decoction On Knee Osteoarthritis In Rat Model

Objective: Through animal experiments,the effect of Bushen Huoxue Decoction on knee osteoarthritis rat model was observed,and the theoretical basis for clinical application of Bushen Huoxue Decoction in the treatment of knee osteoarthritis was provided.Methods:1.Grouping: 30 male SD rats were randomly divided into 3 groups,namely blank group,model group and traditional Chinese medicine group,with 10 rats in each group.2.Modeling: The rats in the model group and the traditional Chinese medicine group were fasted for 12 hours before the experiment,anesthetized by intraperitoneal injection of10% chloral hydrate(0.2ml/100g),and 0.2ml of 4% papain was injected into the joint cavity of both knees.The above operations were performed on the 1st,4th,and 7th days after the start of the experiment.The treatment method of the blank group was the same as that of the model group and the traditional Chinese medicine group.The same amount of0.9% sodium chloride injection was injected into the joint cavity of both knees.3.Administration: After the 10 th day of modeling,gavage was started.The traditional Chinese medicine group was gavaged with Bushen Huoxue decoction 2ml/100 g,and the model group and blank group were gavaged with the same amount of 0.9% sodium chloride injection,twice a day.Continuous gavage for 8 weeks.4.Material collection: After the gavage,the rats were fasted for 12 hours,and were anesthetized by intraperitoneal injection of 10% chloral hydrate.After successful anesthesia,specimens were collected.5.Observation indexes: general condition of rats in each group,serum lipid level,whole blood high shear viscosity,imaging manifestations of knee joints and histopathological morphology of knee joint cartilage.6.Statistical analysis of the experimental data.Results:1.General situation: On the 10 th day of modeling,the rats in the model group and the traditional Chinese medicine group showed decreased spirit and appetite,slow movements,less sensitive response,decreased activity,curled up in a heap,both knee joints were obviously swollen,local bruising,Subcutaneous nodules may even be palpated.After8 weeks of intervention with Bushen Huoxue decoction in the Chinese medicine group,the above symptoms were improved compared with the model group.2.Blood lipid level: Compared with the blank group,the levels of TC,TG and LDL in the model group were significantly increased(P<0.01),and the HDL level was significantly decreased(P<0.01);The levels of TC and TG in rats were significantly decreased(P<0.01),the level of LDL was also decreased(P<0.05),and the level of HDL was significantly increased(P<0.01).3.High shear viscosity: Compared with the blank group,the high shear viscosity of the rats in the model group was significantly increased(P<0.01);compared with the model group,the high shear viscosity of the rats in the Chinese medicine group was significantly decreased(P<0.01).4.X-ray findings of both knee joints: the knee joint space of the rats in the blank group was acceptable,and the articular surface was relatively smooth;the knee joint space of the rats in the model group was narrowed,there were high-density shadows under the cartilage,and the joint surface was frizzy,deformed,and irregular.The boundary is not clear,the osteophytes around the joint are hyperplasia,and intra-articular loose bodies appear.The above-mentioned performances of the rats in the traditional Chinese medicine group were improved.5.HE staining of knee articular cartilage: the cartilage surface of the blank group was smooth,the cartilage layer was thicker,the number of chondrocytes in it was more,the arrangement was neater,and the boundary between the cartilage layer and the subchondral bone was clearer;the cartilage surface of the model group was less regular and the structure After being destroyed,cracks and cavities appear in the cartilage layer,the number of cartilage cells decreases,and they are scattered and arranged in disorder.Compared with the model group,the above-mentioned performances in the traditional Chinese medicine group were alleviated.Conclusion:1.Through this experiment,the knee joint cavity injection papain modelling method can successfully induce the knee osteoarthritis rat model.2.Bushen Huoxue Decoction can down-regulate serum total cholesterol,triglyceride and low-density lipoprotein levels,up-regulate high-density lipoprotein levels,regulate lipid metabolism,and interfere with knee osteoarthritis.3.Bushen Huoxue Decoction can reduce the high shear viscosity of whole blood,reduce blood viscosity,change blood rheology,and play a certain therapeutic effect.4.According to the imaging and HE staining results of this experiment,Bushen Huoxue Decoction can promote the repair of chondrocytes to a certain extent,thereby delaying the progression of knee osteoarthritis.
