Objective Corydalis Rhizoma is a dried tuber of Corydalis yanhusuo W.T.Wang,it has been used for more than 2000 years as a traditional Chinese medicine.Modern pharmacological studies have shown that Corydalis Rhizoma has effect on analgesic,sedative and hypnotic,and it has good effect on many diseases such as coronary heart disease,arrhythmia and gastric ulcer,especially the analgesic effect.In recent years,the formula granules of corydalis have been used in various hospitals at all levels,but in clinical application,they still have many uncertainties in safety,bioequivalence,etc.,and have not yet formed unified standard.Therefore,it has some realistic significance to actively carry out a comparative study on the quality of pieces and granules.Methods In this study,the HPLC fingerprints of Corydalis Rhizoma pieces and its formula granules were established and the main alkaloids were determined,such as protopine,tetrahydropalmatine,corydaline,palmatine,berberine,and then initially clarified the difference of the chemical constituents of Corydalis Rhizoma pieces and its formula granules.On this,acetic acid twisting method and hot plate pain model were used to evaluate the analgesic effect of Corydalis Rhizoma pieces and its formula granules,and to reveal the difference of analgesic efficacy between the two preparations.Finally,the correlation between the chemical composition and analgesic effect was studied by means of gray correlation degree,and the results will provide some theoretical basis for the clinical application of Corydalis Rhizoma pieces and its formula granules.Results In this paper,the HPLC fingerprints of Corydalis Rhizoma pieces and its formula granules was established.There were 15 common peaks in different batches of pieces,and there were 14 common peaks in different batches of formula granules,and the difference between the content of pieces and formula granules was compared by the method of HPLC.The results of analgesic effect showed that the pain threshold of the mice was significantly increased by both the decoction and granule,and the percentage of pain threshold was the most significant in low dose granule.The correlation degree of analgesic spectrum effect is(peak)6>7>5>11>13>15>12>14>2>10>9>3>4.Conclusion According to the analysis results of the fingerprinting of Corydalis Rhizoma pieces and its formula granules,it can be seen that the pieces and formula granules are basically same in chemical composition.The results of analgesic effect showed that the pain threshold of the mice was significantly increased.Analgesia spectrum-effect relationship study showed that the analgesic effect is the result of the combination of protopine,tetrahydropalmatine,corydaline,palmatine,berberine. |
