Objectives1.Study on the fresh-cut processing of Corydalis Rhizoma.2.Study on the equivalence of the fresh-cut slices and the conventional slices.3.To establish quality evaluation system for Corydalis Rhizoma fresh-cut slices。4.To establish qualitative and quantitative model based on near infrared spectrum for fresh-cut slices of Corydalis Rhizoma.Methods1.Investigating the differences of six marker compounds in different specifications,and studying the differences of components through the HPLC fingerprint;Choosing the drying method by investigating the effects of different drying methods on character,extract content and content of tetrahydropalmatine in Corydalis Rhizoma;The hot air drying Kinetics model of Corydalis Rhizoma is established by fitting the change curve of the moisture ratio(MR)during drying process and calculation effective moisture diffusion coefficient(Deff)and the activation energy(Ea);the appropriate range of drying temperature and loading capacity is selected through the single factor investigation,the center combination method is used to optimize the drying process and get the best drying temperature and loading capacity.2.Investigating differences of the fresh-cut slices and the conventional slices in character,identification,total ash and extract;Using HPLC to determinate content of six alkaloids component.Establishing the fingerprint common model of the fresh-cut slices and the conventional slices.Study on the differences of components between the twoby useing the clustering analysis(CA)and principal component analysis(PCA).3.Investigating character,identification,moisture,total ash,extract,multiple marker compounds determination,sulfur dioxide,heavy metal,pesticide residues and aflatoxins of the fresh-cut slices,in order to improve the quality standards of Corydalis Rhizoma.Using high performance liquid chromatography(HPLC)to build multiple marker compounds determination with the chromatographic conditions:the mobile phase is acetonitrile-0.6%acetic acid solution(adjust pH to 5.0 by using triethylamine),gradient elution,the detection wavelength is 280 nm,the column temperature is 25℃,the flow rate is 1.0 mL/min.Using the evaluation system of the similarity of TCM chromatographic fingerprint Version 2.0 to establish characteristic fingerprint of the fresh-cut slices.4.Determining the content of six marker compounds by HPLC,collecting the near infrared spectrum,using partial least squares(PLS)to establish and optimize the multivariate calibration model on correlation of NIR spectrum and HPLC value.Discriminant analysis is adopted to establish and optimize the qualitative identification model.Results1.The content of the second-class is superior to the first-class,the composition is no difference.The content of extract and alkaloid composition have no differences,but the character has.So 4mm is chosen as the cut thickness.Corydalis Rhizoma slices which use hot air drying process,the character is good,the highest extract content,content of alkaloid components is higher,so the hot air drying process is chosen as origin drying process of Corydalis Rhizoma fresh-cut slices.Using Modified Page model to build MR-t function,it can accurate predict moisture ratio curve under different temperature and loading capacity,effective moisture diffusion coefficient is 3.972 × 10-10~1.362×10-9 m2/s,the activation energy is 30.17 kJ/mol.The regression equation was established by the response surface optimization experiment.The optimum drying process was determined as the slice thickness 4mm,the loading capacity was 7kg/m2,the drying temperature was 85℃,the total drying time was 279.0 ± 1.1 min,and the comprehensive score was 0.8606 ± 0.01 by calculating and combining with the actual production.2.The fresh-cut slices is better than the conventional slices in terms of characters,microscopic structure.there is no difference in terms of extract content.the conventional slices is better than the fresh-cut slices in terms of total ash content.Through T test analysis,there are significant differences berween the fresh-cut slices and the conventional slices on content of six alkaloids component.By comparing similarity of fingerprint,there are no differences in composition of component between the two.Through cluster analysis,the differences between the two is obviously.Through principal component analysis,protopine,palmatine,berberine,dehydrocorydaline,tetrahydropalmatine,corydaline.are characteristic alkaloids component in Corydalis Rhizoma.Fresh-cut slices is superior to the conventional slices in the component content3.The comprehensive quality evaluation system of Corydalis Rhizoma fresh-cut slices is established,including character,identification,examination,extract,and determination of sulfur dioxide,heavy metals,pesticide residues,aflatoxin.Established the method to determine content of six effective composition in Corydalis Rhizoma.Established fingerprint common mode of Corydalis Rhizoma fresh-cut slices,similarity of spectrum and fingerprint is above 0.95,there are 25 common chromatographic peak,among which,No.7,No.13,No.15,No.16,No.18,No.22 is protopine,palmatine,berberine,dehydrocorydaline,tetrahydropalmatine,corydaline.4.The model was established through PLS,selecting waveband of 8770.32~4242.63 cm-1,using "MSC+1 stD" as pretreatment method to optimize model,get quantitative detection model of six marker compounds in Corydalis Rhizoma fresh-cut slices,model prediction accuracy is higher by validating the prediction set.The model was established through discriminant analysis,selecting waveband of 8770.32~4242.63 cm-1,using "SNV+1 stD+Nd" as pretreatment method to optimize model,get qualitative identification model Corydalis Rhizoma fresh-cut slices,model prediction is correct by validating the prediction set.Conclusion1.Established fresh-cut processing technology of Corydalis Rhizoma,provided a preliminary basis for pilot test.2.The fresh-cut slices is superior to the conventional slices in term of character and composition,fresh-cut processing can improve the quality of Corydalis Rhizoma3.The comprehensive quality evaluation system of Corydalis Rhizoma fresh-cut slices can be very good control the quality of the fresh-cut slices4.The NIR qualitative and quantitative correction model can quickly and accurately identify two kinds of slices and preliminary predict sixe alkaloids content...
