| Objective:In initial study,our research team used neurophilic virus tracing technology to screen Feishu points and lower respiratory tract related brain nuclei,and the results suggested that the Edinger-Westphal nucleus(EW nucleus)was related with Feishu point and lower respiratory tract.Using microdialysis and liquid-mass spectrometry techniques to analyze the effect of acupuncture on the neurotransmitter of the EW nucleus of asthmatic rats,the results suggest that the change of glutamate content in the EW nucleus is related to the regulation of respiratory function by acupuncture at Feishu point.The neurobiological mechanism of the EW nucleus nerve on the regulation of respiratory function and the effect of acupuncture at Feishu point remains to be studied.Method:In this study,we use normal rats as the research platform to observe the regulation of the EW nucleus on the respiratory function by electrical stimulation of nucleus experiment,the multi-channel in vivo recording technology to analysis the effect of methacholine(Mch)aerosol attack and acupuncture at Feishu point on the EW nucleus neuronal firing.Results:1)The frequency of rat breathing enhancement waves was significantly increased after electrical stimulation of EW nucleus and NTS nucleus(P<0.01).After 3 times electrical stimulation of NTS nucleus,the number of respiratory enhancement waves showed an increasing trend;while 3 times electrical stimulation of EW nucleus were no difference between each stimulation;After the electrical stimulation of the NTS nucleus,the arterial blood pressure decreased significantly(P<0.01),and the blood pressure decreased after each electric shock(P<0.05),and there was no significant change in arterial blood pressure after electrical stimulation of EW nucleus(P>0.05).2)After Mch attack,the EW nucleus and NTS nucleus neurons are mainly excitatory firing.In the EW-Mch group,the firing rate of the EW nucleus reached the highest peak within 300 s after the nebulization of Mch attack,and then the neuronal firing rate gradually decreased.The neuronal firing rate of NTS nucleus neurons in the NTS-Mch group reached the highest peak within 150 s after the nebulization of Mch attack,and then tended to rise gradually.3)The number of activated neurons in the EW nucleus and NTS nucleus of acupuncture at the ipsilateral acupoint was significantly higher than that of acupuncture at the contralateral acupoint(P<0.01);the number of activated EW nucleus and NTS nucleus neurons after acupuncture at Feishu acupoint was significantly higher than that of acupuncture at Chize acupoint and Zusanli acupoint(P<0.01),and the number of activated neurons in the EW nucleus of acupuncture at Feishu acupoint is higher than that of NTS nucleus(P<0.05);acupuncture at the ipsilateral Feishu acupoint is associated with continuous excitability,compared with the types of transient excitability firing,transient inhibitory firing,and continuous inhibitory firing,the EW nucleus has the highest change in the neuronal firing rate of the excitatory neuron(P<0.05);during the sequential needle insertion and twisting operations,the EW nucleus is instantly excited,the neuronal firing rate of excitatory neurons has a trend of increasing.4)Acupuncture at Feishu and nebulization of Mch attack both activated EW nucleus neurons,but the proportion of EW nucleus inhibitory neurons activated by acupuncture at Feishu(21%)was higher than attack of Mch nebulization(4%).Conclusion:1)The EW nucleus is related to respiratory rhythm and inspiratory function,and can enhance the frequency of active breathing.This function has no difference with arterial blood pressure.2)Changes in respiratory function affect the activity of EW nucleus and NTS nucleus neurons;the response time of EW nucleus neurons to changes in respiratory function is later than NTS nucleus and the duration is shorter than NTS nucleus.3)The EW nucleus has a specific response to acupuncture at Feishu acupoint on both sides,but it has more significantly response to acupuncture at ipsilateral Feishu acupoint.4)The response pattern of EW nucleus is different between to changes in lung function and acupuncture at Feishu acupoint. |
