| Objective:The difference of NOP16 expression level in lung adenocarcinoma tissue and lung normal tissue was analyzed,and the relationship between different NOP16 expression levels and the clinical characteristics and prognosis of patients was compared,and the correlation between NOP16 and immunity in lung adenocarcinoma was further analyzed.Methods:The NOP16 m RNA sequencing data and clinical information data of lung adenocarcinoma were obtained through the TCGA database.The differential analysis of NOP16 expression in lung adenocarcinoma and normal tissue was carried out,and the relationship between different expression levels of NOP16 and clinical characteristics was compared.Independent prognostic analysis was used to evaluate whether NOP16 acts as an independent prognostic factor.Gene enrichment analysis explores the possible pathogenesis of NOP16 in lung adenocarcinoma.Then the relationship between NOP16 and immune cell infiltration and tumor microenvironment score was analyzed.Survival analysis was used to evaluate the impact of high and low expression levels of NOP16 on prognosis and the impact of different tumor microenvironment scores on prognosis.Results:The expression level of NOP16 in lung adenocarcinoma tissue was significantly higher than that in normal lung tissue(P<0.001),and the high expression of NOP16 was significantly correlated with tumor lymph node metastasis and pathological stage(P<0.05).NOP16 survival analysis showed that those with high expression of NOP16 had worse prognosis(P=0.005).Independent prognostic analysis showed that NOP16(HR=1.371,95%CI=1.053-1.785,P=0.019)and pathological stage(Stage II,HR=2.168,95%CI=1.454~3.234,P<0.001;Stage III,HR=2.820,95%CI=1.870~4.254,P<0.001,Stage IV,HR=3.188,95%CI=1.711~5.942,P<0.001)can be used as an independent prognostic factor.GO analysis showed that the cellular components involved in NOP16 include ribosomes,molecular chaperone complexes,mitochondrial matrix,oxidoreductase complexes,and peptidase complexes;molecular functions include r RNA binding,unfolded protein binding,and ribonucleoprotein complex binding,translation factor activity RNA binding,RNA methyltransferase activity;biological processes include ribosome biogenesis,r RNA metabolic processes,ribonucleoprotein complex biogenesis,non-coding RNA processing,mitochondrial translation.KEGG results showed that the high expression of NOP16 was enriched in ribosome,spliceosome,fluorouracil metabolism,oxidative phosphorylation and other pathways(P<0.05).In addition,the expression level of NOP16 was significantly correlated with immune cells such as B cells,neutrophils,CD4+T cells,CD8+T cells,macrophages,dendritic cell infiltration,and myeloid-derived suppressor cells(all P<0.01)and There were significant correlations with tumor microenvironment scores such as immune score,stromal score,Estimate score,and tumor purity(all P<0.01),and different tumor microenvironment scores had a significant impact on prognosis(P<0.05).Conclusion:NOP16 is highly expressed in lung adenocarcinoma,and it is associated with poor prognosis,immune cell infiltration,and tumor microenvironment score in lung adenocarcinoma patients,and may serve as a potential therapeutic target. |
