| Objective: Core-Binding Factor Runt Domain Alpha Subunit 2 Translocated To 3(CBFA2T3)was evaluated and analyzed by bioinformatics to investigate the expression difference of CBFA2T3 in lung adenocarcinoma(LUAD)and its significance as a brand-new potential prognostic biomarker for LUAD,to study the effect of CBFA2T3 on immune infiltration in LUAD,and to further analyze the KEGG pathway and biological function involved in its co-expression of highly related genes.Methods: The expression differences of CBFA2T3 in pan-cancer were compared by GEPIA2(Gene expression Profiling Interactive Analysis 2)database;the prognostic value of CBFA2T3 in pan-cancer was assessed by GEPIA2 and KM(Kaplan-Meier Plotter database)databases;the relationship between CBFA2T3 and immune cell invasion,tumor clinical stage and related gene markers was analyzed using TISIDB,GEPIA2,and TIMER(Tumor Immune Estimation Resource)databases.The possible involvement of CBFA2T3 in LUAD was elucidated by GO and KEGG enrichment analysis.CBFA2T3 associated mi RNAs were screened and predicted using the mi Rwalk database.Results: CBFA2T3 expression is decreased in most malignancies and is associated with poor prognosis in LUAD.CBFA2T3 was highly correlated with the prognosis of patients with LUAD with different clinical parameters,and significantly affected the survival time of LUAD patients.CBFA2T3 expression was positively correlated with functional T-cell infiltration,involving exhausted T cells and other immune cell infiltration.In addition,CBFA2T3 interacts with TIGIT,LAG3,CXCL13,LAYN,GZMB,TIM-3,and PD-1,key gene markers of exhausted T cells.Functional enrichment analysis showed that CBFA2T3 may be involved in adaptive immune response,regulation of leukocyte activation,related to leukocyte differentiation,and involved in cell chemo-taxis and immune response regulation signaling pathways.A mi RNA-gene network consisting of CBFA2T3 and 375 mi RNAs was constructed by screening and predicting mi RNAs that regulate CBFA2T3.Conclusions: CBFA2T3 is a potential diagnostic and prognostic biomarker in LUAD,and CBFA2T3 is an important factor in promoting T cell failure in LUAD and is associated with immune infiltration in LUAD.Detecting CBFA2T3 expression may help predict the prognosis of lung adenocarcinoma,and regulating CBFA2T3 expression in exhausted T cells may provide a new therapeutic strategy,thus improving immunotherapy for LUAD. |
